Allium victorialis L.is a perennial herb of Liliaceae.It has the functions of hemostasis,chemophilia,expectoration,pain relief,cancer prevention and so on.Allium victorialis L.is a good plant for food and medicine.Seeds are difficult to break the dormancy.So that has low propagation coefficient and a long cycle of seed propagation.The rapid propagation of Allium victorialis L.is based on tissue culture technology.In this experiment,Allium victorialis L.was used as experimental materials,screening the basic medium and exogenous substances to obtain regenerated plants.Then the rapid propagation system of tissue culture was established.The results showed that when the final concentration of 6-BA was 2.0 mg/L and the final concentration of NAA was 1.0 mg/L,the induction rate of cold onion bud was up to 97% when the final concentration of 6-BA was 2.0 mg/L and NAA was 1.0 mg/L in MS medium.When NAA with final concentration of 3.0 mg/L was added to MS medium,the effect of inducing proliferation and subculture was the best.The results showed that the addition of 2.0 mg/L IBA to MS medium could effectively promote the root growth of it.The results showed that when the final concentration of 6-BA was 1.0 mg/L and the final concentration of 0.1 mg/L NAA was added to the MS medium,it was the best condition.The optimum culture conditions for adventitious buds showed that the induction rate of adventitious buds was up to 75.6% when the medium was 6-BA 0.5 mg/L NAA 0.01 mg/L KT 0.1 mg/L.The best culture conditions for rooting were also selected.The results showed that the rooting rate of MS medium was 87.2% when the final concentration of NAA was 0.02 mg/L.Finally,the callus culture was carried out with the root cap of Allium leek as explants.The optimum culture conditions were as follows: 1.0 mg/L 6-BA and 0.1 mg/L NAA.in MS medium.The optimum culture conditions of meristem and leaf and rooting were optimized.The results showed that the optimum culture conditions of meristem and leaf were 6-BA of 2.0 mg/L and NAA.of 0.1 mg/L in MS medium.The best rooting culture condition was that NAA.of 0.1 mg/L was added to MS medium..In this study,the callus induction and regeneration system with stem,leaf and root cap of Allium chives as explants was established,which laid a good foundation for the establishment of genetic transformation system of Allium victorialis L.Provide technical support with higher efficiency and low cost.
