In order to accurately determine the dietary fiber content in soybean seeds and analyze the genetic mechanism of soybean dietary fiber related traits.In this study,the national standard method was used to determine the accurate method for the determination of dietary fiber in soybean seeds by using single factor analysis and response surface analysis,and the genetic population of jinda 53 × pingnan was determined by this method,and the gene location was analyzed.The main research results are as follows:(1)Experiments have found that there are three factors that affect the accurate determination of soybean dietary fiber: fat,starch,and protein.Different size of soybean meal and different degreasing time have an effect on the degreasing effect.Through 100 sieve bean meal,only 6 H can achieve the effect of complete degreasing;When the substrate mass is 0.5 g,the amylase addition amount is 4500 μL,the amylase hydrolysis time is 20 min,the glycosase addition amount is 500 μL,and the glycosase enzymatic hydrolysis time is 40 min,and the amylase hydrolysis is complete;The protease addition amount is 36,900μL and the enzymatic hydrolysis time is 35 min,which can maximize the enzymatic hydrolysis effect of the protein.(2)Because different dietary fiber components have different physiological functions for the human body,the fiber quality related traits of genetic groups are first cluster analyzed,and the content of fiber components of each group is different,providing a basis for the subsequent selection of high-quality germplasm.Secondly,by locating the related traits of soybean seed dietary fiber,it was found that insoluble dietary fiber,acid washing fiber,and lignin were all labeled in the range of 155.92-156.32 c M in the 5th chain group.Using the sequence information of the positioning area,the results of the annotation of soybean grain fiber quality gene were analyzed.It was found that the area containing 0.4 c M contained a total of 4 candidate genes related to sugar metabolism,namely PACid: 26320833,PACid: 26319674,PACid:26319675,PACid: 2632581.Therefore,the four genes were considered as the most likely candidate genes for soybean grain fiber quality gene.(3)First of all,the fiber quality related traits of the capsule of the genetic population were cluster analyzed.The fiber component content of each group was different,providing a basis for the subsequentselection of high-quality feed germplasm.Secondly,through the QTL to locate the related traits of soybean capsule fiber,it was found that the loci effect of pod hemicellulose was the strongest in the 15 th chain group,and the explicable genetic variation was 11.21%,which was presumed to be the main gene.Acid washing fibers and hemicellulose in the pods ’skins were marked in the 76.13-79.41 c M range of the 15 th chain group.A gene related to sugar metabolism was found by analyzing the results of gene annotation(PACid: 26329810).Therefore,the gene was considered as the most likely candidate gene for soybean grain fiber quality gene.
