Cotton is an important economic crop with a very obvious heterosis.However,the research and utilization of cotton heterosis were later than other crops.Cytoplasmic male sterile was the basis of the heterosis utilization,and its mechanism need to be further studied because of its complex.In this study,in order to expole the biological function of Gh GA20ox1 gene during the period of the plant growth and development,CMS(cytoplasmic male sterile)Jin A and its maintainer were used as materials,Gh GA20ox1 gene was cloned,analyzed by bioinformatics,and contrasted between Jin A and its maintaine,the result bring the basis not only for the relative study between Gh GA20ox1 gene and CMS but also for exploring the Gh GA20ox1 gene regulation mechanism to cotton anther.The results were as follows:1.The full-length c DNA of cotton Gh GA20ox1 gene and genomic DNA were cloned using the method of homologous cloning,and analyzed by bioinformatics.The c DNA of Gh GA20ox1 gene from Jin A sterile line and its maintainer line were all 1107 bp,encoding 368 aa.The full length of the genomic DNA of Jin A and its maintainer line was 1707 bp,which consisted of 3 exons and 2 introns.The multi-sequence alignment of the protein revealed the conservation of Gh GA20ox1 proteins were general,they were only conservative within a conserved domain.2.The expression of Gh GA20ox1 gene by the q RT-PCR between Jin A sterile line and its maintainer line during different stages of microspores aborted.The result showed that the expression of GA20ox1 gene from the maintainer line were consistent during the different stages.the expression of GA20ox1 gene from Jin A were significantly lower than that of maintainer line between the stages before microspores abortion and the microspores abortion.Because the regulation of GA to development of stamens was achieved by regulating tapetum and developing pollen.So it is speculated that the reason of the microspores abortion of the Jin A sterile line is the reduction of GA content because of the down expression of Gh GA20ox1 gene.The dysplasia of the tapetum development due to the GA signal change cause the microspores abortion.
