Effects Of Antioxidants And Sugar On The Quality Of Cooled Stored And Frozen-thawed Equine Semen

In this study,pure blood horse semen was used as the main research object,and saccharides and antioxidants were added to semen.In order to explore its effect on low temperature and cryopreservation of horse spermatozoa,the kinetic parameters were detected by sperm analyzer and the plasma membrane integrity and mitochondrial membrane potential were measured by fluorescence staining.We prepared horse semen dilutions and analysed the differences of semen quality in different dilutions by adding different saccharides and their combination to the basal diluent,the following conclusions can be drawn.1: Semen diluent prepared by adding only raffinose,lactose or raffinose or lactose to semen diluent is not suitable for cryopreservation of horse semen;2: The semen dilutions prepared by different carbohydrate combinations failed to achieve the low temperature preservation and cryopreservation effect of semen which was superior to the classic glucose dilution dilution of glucose + lactose.We prepared horse semen dilutions and analysed the differences of semen quality in different dilutions by adding different antioxidantand their combination to the basal diluent,the following conclusions can be drawn.1: Semen diluent prepared by adding vitamin C or taurine has better semen quality at low temperature preservation;2: The semen diluent prepared by adding methionine can maintain the mitochondrial membrane potential and plasma membrane integrity of spermatozoa,and the survival time of spermatozoa after freezing thawing can be prolonged appropriately;3: When the concentration of cysteine was 2.5 mmol/L,the prepared semen diluent decreased the sperm quality of low temperature and cryopreservation,which was not good for sperm preservation.
