Potato(Solanum tuberosum L.)is an important crop with high yield and adaptation to diverse environmental conditions.The tuber of potato is not only an economic organ but also a reproductive organ.Studying the mechanism of tuberlization provides a guide of improving the yield and quality.The photoperiod-respond CO-FT pathway regulates multiple developmental processes such as flowering,seasonal growth,and storage organ formation.The FT homologous gene St SP6 A mediates photoperiodic signals and regulates tuberlization.However,it is unclear that how St SP6 A further regulates the expansion of the tip of the stolon.Based on the conserved FT-AP1 regulatory module in the apical meristem,this study identified all of MADS-box genes in the whole potato genome and clarified its gene structure and expression patterns under different photoperiod conditions.Screening MADS-box transcription factors that may be involved in the regulation of photoperiod-induced storage organ formation by St SP6 A signaling can lay the foundation for investigation of signal transduction of St SP6 A.The major results are as follows:1)132 MADS-box transcription factors in potato were identified and analyzed for clustering,chromosomal location and gene structure by utilizing the plant TFDB database.The results showed that MADS-box transcription factors were clustered in two subfamilies: MIKC-type and M-type.M-type was further divided into three subfamilies: M?,M? and M?;MADS-box has similar genetic structure in the subfamily,and it is unevenly distributed on the 12 chromosomes of potato with the phenomenon of a clustered distribution.2)Through the analysis of the expression data of DM1-3 and RH which is published in the potato genome database spud DB,56 MADS-box transcription factors that expressed in stolons or tubers were screened.24 MADS-box transcription factors among them differentially expressed in four stages of tuberlization were chosen.And the 24 MADS-box transcription factors were further analyzed under different photoperiod conditions.We found that 10 MADS-box transcription factors differentially expressed under short day.10 MADS-boxes were further verified in The phytochrome F interference plant,and the results were consistent with those of photoperiod.3)The promoter cloning analysis of the 10 MADS-box genes we obtained showed that all of them contained the b ZIP family transcription factor core binding element.The cis-elements predicting results of the 10 MADS-box genes? promoter regions showed that there were differences among diverse MADS-box functional elements,but they all have their own light responsive elements.4)The TAC does activate the expression of five AP1 like genes and was confirmed by Agrobacterium-mediated transient expression of Nicotiana benthamiana;The subcellular localization results of DMT63313 and DMT68325 showed that they were localized in the nucleus.
