Rice(Oryza sativa.L)is the principal crop in the world and it is particularly important for the food security.Salt stress has a great influence rice yield in agriculture.Salt stress can significantly affect the growth and development of rice,causing abnormal physiological and biochemical reactions in rice,which can result in a significant reduction in yield.Therefore,studying the mechanism of salt tolerance in rice and screening rice salt-tolerant genes are crucial for crop breeding for cultivating high-quality and high-yielding rice.In this study,we characterized three genes,which regulate salt stress on rice,and provided a reference for the study aimed at rice salt tolerance mechanism.Functional studies of Os53(LOC_Os02g53110):Os53 is also a typical DUF1644 family of protein with a DUF1644 conserved domain and a zinc finger domain.We confirmed that O.s53 located in the nuclear and cytoplasm by subcellular localization.GUS staining results indicated that the protein was constitutive expression.We constructed overexpression(Os53OE)and RNAi transgenetic plants(Os53RNAt)of Os53.We found that Os53 negatively regulate the salt tolerance of rice.Functional analysis of Os23(LOC Os07g23 790):Os23 is a member of the DUF1644 family with a typical DUF1644 family conserved domain and zinc finger domain.Subcellular localization revealed that Os23 localized in the nucleus.qPCR revealed that O.s23 expressed in various growth and development stages of rice and belonged to constitutive expression.After treatment with abiotic stress,Os23 was involved in the regulation of rice resistance to salt stress.Overexpression of Os23(Os23OE)significantly reduced the ability of rice to resist salt stress.When we knocked down the expression level of Os23(Os23RNA),it resulted in that the ability of rice to resist salt stress was significantly improved.Therefore,we concluded that Os23 negatively regulates rice resistance to salt stress.Os16(LOC_Os03g16460)functional study:Os 16 is a protein that we screened by yeast two-hybrid assay and interacts with the DUF1644 family of protein Os05.Our study found that Os 16 has a similar function to the DUF1644 family of proteins and plays an important regulatory role in abiotic stress in rice.We found that the Osl6 localized in the nucleus and cytoplasm.qPCR expression patterns analysis showed that Osl6 was expressed in various growth stages of rice and belonged to constitutive expression.At the same time,we found that in overexpressing Os 16 transgenitic plants(Os16OE),the ability of rice to resist salt stress significantly improved.
