The roots and rhizomes of Valeriana officinalis are used as medicines for calming and soothing,relieving pain,anti-depression and anti-tumor,increasing coronary flow,anti-arrhythmia,gastrointestinal smooth muscle spasm and other pharmacological activities.As people’s life pressures,insomnia,depression,cancer and other diseases were increasingly plaguing human health.The demand for valerian extract and its preparations is increasing.However,the natural growth of valerian was very slow,the wild valerian resources are drastically reduced,and artificial cultivation was difficult.Not only the root growth was slow,but also the genetic stability was difficult to maintain,which greatly affects its quality and yield.Therefore,the use of V.officinalis roots as raw materials for medicinal materials was an effective means to solve the problem of insufficient supply of valerian extract and its preparations.In this study,the seeds of V.officinalis were used as the material to culture sterile seedlings,and the stems and leaves of the sterile seedlings were used as explants to induce adventitious roots.After subculture and proliferation,the adventitious roots with uniform thickness were selected to explore the hormone concentration and type,medium type,pH,and sugar source.The effects of species,sugar concentration,salt concentration,total N concentration and total P concentration on the suspension culture of adventitious roots.The optimal conditions for the suspension culture of adventitious roots were established.The system of high-efficiency culture of adventitious roots was preliminarily established,and the growth status,the content of valepotriate,acevalepotriate and valerenic acid of adventitious roots in this system were investigated.As a result,the high-yield adventitious roots provided a theoretical basis to further cultivated by bioreactor.Experimental results showed:1.In the combination of five kinds of disinfectants and three kinds of immersion disinfection time,soaking for ten minutes with 4%NaCIO solution,the seed was non-pollution and the germination rate was highest reaching 76%.2.The ideal conditions of the adventitious roots induction V.officinalis was that:the explant was stem segments and the medium was MS+0.5 mg-L-1 NAA.3.The ideal culture conditions for the suspension culture of adventitious roots of Vofficinalis were followed:the basic medium was twice of major element concentration,48.46 mmol·L-1 total N concentration,1.5 mmol L-1 total P concentration of RCM,and added sucrose 40 g·L-1,added NAA 0.5 mg·L-1,and adjusted pH at 5.7.Finally,harvest or subculture at 28 d after inoculation.4.It was also found that The contents of the three components in the normal adventitious roots cultured under the ideal system were higher,the content of valepotriate was 7.222 mg·g-1,the content of acevalepotriate was 0.355 mg·g-1,the content of valerenic acid was 4.016 mg·g-1,and the content of valerenic acid was greater than the two-year-old seedling roots.
