Selenium and alfalfa play an important role in regulating the immune function of the body Selenium deficiency can cause 1g synthesis and lymphocyte proliferation and differentiation.Excessive supplementation of selenium can cause cellular immunity and humoral immunity.The application of alfalfa is more ideal when the growth cycle is longEarlier,we found that the combination of yeast selenium and alalotin can significantly increase the content of egg selenium,the laying rate of laying hens and the antioxidant capacity of the body,but the effect of the combination of the selenium and alalotin on the immune effect of the animal body has not been reported.Therefore,on the basis of previous studies,the present study investigated the effects of the combination of yeast selenium and rubidin on apoptosis and immune function in the liver and spleen of laying hens,aiming at improving the immune performance of laying hens in the post-laying period.Providing Theoretical Basis and Technical Support for Sustainable Development of Layer Industry.69-week-old 432 healthy Henlan Brown laying hens were randomly divided into 4 treatment groups,each containing 6 replicates and 18 chickens per replicate.In the control group,the basal diet was fed on the basal diet.The experiment group added 1000mg/kg alalice and different doses of yeast selenium(0.2,0.6,1.0mg/kg)in the basal diet for 2 weeks,and the test period was 8 weeks.The effect of HE staining,TUNEL,ELISA analysis and real-time fluorescence quantitative PCR on the apoptosis and immune function of the liver and spleen cells of egg laying hens at the late stage of laying eggs was studied.The results show that:(1)HE staining showed that the combination of yeast selenium and alalotin could improve the liver fatty degeneration,the disorder of the hepatic cord,improve the massive hemorrhage of the spleen,the red pulp and the white pulp,and the height of the jejunum,the value of V/C(P<0.05),and the depth of the recess(P<0.05)(2)The combination of yeast selenium and alfalfa had no significant effect on serum IgA,IgG and IgM(P>0.05)(3)The combination of yeast selenium and alfalfa can significantly reduce the apoptosis rate and the expression of Caspase-3 mRNA in liver and spleen of laying hens(P<0.05)4)The expression of TNF-a,NF-kB,IL-6 mRNA and protein in the liver and spleen of layer chickens were not significantly different(P>0.05),but there was a decreasing trend;In 0.6 mg/kg yeast seleniumx 1000 mg/kg oryzae,the expression of IL-2 mRNA in liver increased significantly(P<0.01),protein expression was significantly increased(P<0.05),IL-2 mRNA expression in spleen and protein The expression level significantly increased(P<0.01);when 0.2 mg/kg yeast selenium and 1000 mg/kg were added,the expression of IL-4 mRNA in the liver was significantly increased(P<0.05),and the expression of mRNA in the spleen was significantly increased.Increased(P<0.05),protein expression levels increased significantly(P<0.01).In summary,the addition of yeast selenium and luteolin to the diet can reduce the liver and spleen damage and apoptosis rate,inhibit the expression of pro-apoptotic and proinflammatory cytokines,increase the production of anti-inflammatory factors,and enhance the body.Immune Function.Based on yeast selenium 0.2 mg/kg basal diet,the combination of 1000 mg/kg luteolin and 0.2-0.6 mg/kg yeast selenium was better.The combination is better.
