This paper used two advanced seed treatment equipment,plasma and magnetic fields to treat the Sophora flavescens,in order to solve the germination potential,low germination rate,unsettled emergence and other problems in the production practice of Sophora flavescens,and to comeover various shortcomings in the treatment of seeds in production practice.The seed treatment equipment is used to treat the seeds of Sophora flavescens,in order to find the optimum conditions suitable for the germination and growth of Sophora flavescens,and to explore the mechanism of action of the two physical methods,Thus,it lays a foundation for the study of seed action mechanism by production practice and two physical methods.(1)After different strength magnetic fields and plasma treatment of Sophora flavescens seeds,their biological effects showed great differences.425 GS(25.0 A)magnetic field treatment of Sophora flavescens seeds for 30 minutes or 2.0 A plasma treatment of Sophora flavescens seeds for 3.0s,the germination potential,germination rate,root length,plant height,number of lateral roots and the Fresh weight roots of Sophora flavescens is the best.For the content of oxymatrine in the roots of Sophora flavescens,the magnetic field strength is in the range of 12.5 A-37.5 A(209 GS-611 GS),which is beneficial to the synthesis of oxymatrine in the roots of Sophora flavescens.The intensity field of 50.0 A-62.5 A(835 GS – 1016 GS)will inhibit the synthesis of oxymatrine in Sophora flavescens.With the increase of plasma strength,the content of oxymatrine decreased first and then increased,and the highest content of oxymatrine is 3.0 A.After magnetic field or plasma treatment of Sophora flavescens seeds,the placement time has a great influence on the effect of the magnetic field or plasma to promote the seeds of germination.The results showed that the magnetic field had the best effect on the germination of Sophora flavescens.When the seeds of Sophora flavescens were placed 9 d-12 d after magnetic field treatment,and when the plasma was placed at 6 d after treatment,the germination of Sophora flavescens was the most favorable.After magnetic field or plasma treatment of Sophora flavescens seeds,the placement time has a great influence on the effect of the magnetic field or plasma to promote the seeds of germination.The results showed that the magnetic field had the best effect on the germination of Sophora flavescens when the seeds of Sophora flavescens were placed 9-12 d after magnetic field treatment,and when the plasma was placed at 6 d after treatment,the germination of Sophora flavescens was the most favorable.To research the magnetic field and plasma to promote the germination of Sophora flavescens seeds,mainly from four aspects: the submicroscopic structure of seed skin,respiration rate,swelling rate and conductivity of seed leaching solution.The results showed that after the magnetic field treatment,the epidermis scratches of the seeds of Sophora flavescens increased,the residual tissue of the umbilical part reduced,the imbibition and respiration rate of the seeds increased,and the conductivity of the seed leaching solution decreased.All four kinds of changes could stimulate its germination and change.(2)The best hormone combination for seed mature embryo-induced callus was 1.0 mg/L 2,4-D+2.5 mg/L 6-BA.Under the intensity of 425 GS magnetic field,different treatment times had different effects on the growth of callus and the content of oxymatrine in its internal oxidation.The callus of Sophora flavescens treated with a magnetic field of 25.0 A(425 GS)for 30 min can promote the growth of callus and increase the content of oxymatrine.(3)The magnetic field has a certain stimulating effect on the germinated seeds.After the magnetic field treatment,the change of the root length and the number of lateral roots is the most obvious.Under 25.0A(425GS),the optimal treatment time was 30 min.And under 37.5 A,the best treatment time is 20 min.(4)Under drought stress,the drought resistance of Sophora flavescens seeds which treated by magnetic field and plasma was improved.The drought resistance of Sophora flavescens is the best when the magnetic field treatment strength is 25.0 A(425 GS)or the plasma intensity is 1.5 A-2.5 A.The SOD and POD activities of Sophora flavescens seeds were significantly increased by different strength of magnetic fields.At the same time,the content of MDA was reduced,which finally made the Sophora flavescens better suitable for drought stress and improved its drought resistance.After treated by plasma,the activity of POD in the roots of Sophora flavescens was not obvious changed under drought stress,but because of the activity of SOD was significantly increased and the content of MDA was decreased.Finally the drought resistance of Sophora flavescens was improved.
