As a kind of staple food for the most of human beings all over the world,the nutritional value of rice is attracting more and more attention.Rice contains a lot of nutrients,among which vitamins,amino acids,lipids,flavonoids and so on are the focus of people's attention.These nutrients play an important role in human life.For example,vitamins are involved in many metabolisms as cofactors.However,the types and contents of these nutrients are quite different in different varieties of rice.Therefore,it is particularly important to select high-quality varieties by comparing the differences of nutrients in different varieties.In this study,the contents of various nutrients in 533 core germplasm and 5 special rice varieties were compared by metabolomics method,and high-quality varieties were selected for breeding.First of all,based on our determined metabolite,we has broad target to measure the17 kinds of amino acids,67 species flavonoids,16 kinds of lipid and the material content of 8 kinds of vitamins in 533 core germplasm.we found that the amino acids,lipids,vitamins and other primary metabolites are abundant in 533 core germplasm,at the same time,we also found the secondary metabolites flavonoids substances are in the potential of natural variation in groups.PCA showed that flavonoids was obvious different in indica and japonica.We did cluster analysis on the substance content of each substance in the population,and combined with the line chart of the substance content with the largest variation of each nutrient,we selected varieties R319 as high flavonoid content,varieties R516 and R477 as high lipid content,varieties R516 and R518 with high amino acid content,and varieties R319 with high vitamin content.Then,these high-quality varieties with five other special rice varieties are compared.the results showed that R477,R518 and JZ are significantly higher content of amino acids in seven other varieties,R477 and JZ are significantly higher content of section of lipid in the seed higher than the other eight varieties.Content of flavonoids in R477 and HX is significantly higher than the other eight varieties,R477's vitamin content in the seed is significantly higher than the other nine varieties.By selecting the optimal varieties,each of these varieties were used for breeding work in the future.Secondly,many genes controlling flavonoid content,including many synthetic genes and transcription factors,were located by genome-wide association analysis,laying a foundation for the exploration of genetic variation of flavonoids.In summary,through the analysis and comparison of nutrients in different varieties,we selected the high-content varieties corresponding to each nutrient,and studied the natural variation of flavonoids in 533 core germplasm,which can provide methods and theoretical references for the breeding of new nutrient-type rice varieties in the future.
