In livestock production,skeletal muscle growth and quality traits is important for the meat performance evaluation indicators of food-producing animals.Non-coding genes,such as microRNA and lncRNA,which have been widely studied,play an important role in the regulatory network of skeletal muscle development.Our laboratory has previously found that long non-coding RNA SYISL can promote the proliferation of myoblasts and inhibit the differentiation of myoblasts.In this study,we used bioinformatics analysis to predict that miR-34 b could bind to SYISL,and further studies found that miR-34 b could play a regulatory role in myogenesis by targeting SYISL.The specific research results are as follows:1.q-PCR is used to detect the expression of miR-34 b in different tissues of mouse,the results showed that miR-34 b is highly expressed in the leg muscles,and its expression level increased gradually during the differentiation of C2C12 cells.q-PCR,Western Blot and immunofluorescence are used to detect the related genes of myoblast proliferation and differentiation after miR-34 b inhibition and overexpression,the results showed that miR-34 b inhibits myoblast proliferation and promotes myoblast differentiation.2.Bioinformatics analysis,Dual-Luciferase reporter gene system,RNA pull-down and RNA Binding Protein Immunoprecipitation are used to prove that miR-34 b can directly bind to SYISL and AGO2 protein.3.q-PCR results showed that miR-34 b reduce the expression of SYISL and co-transfection experiment results showed that miR-34 b regulates the proliferation and differentiation of myoblast by targeting SYISL.4.q-PCR and Western Blot results showed that miR-34 b can promote the expression of SYNPO2,which is the flanking host genes of SYISL.In conclusion,miR-34 b inhibites the proliferation of myoblasts and promotes the differentiation of myoblast,and miR-34 b regulates the proliferation and differentiation of myoblasts by targeting SYISL.These results provide a new regulatory mechanism for skeletal muscle growth and development,and have important significance for further understanding the molecular regulation of skeletal muscle growth and development.
