In order to investigate the effect of Clostridium butyricum on the fermentation quality and nutrient content and its aerobic stability of high moisture corn silage.By means of morphological features,physiological and biochemical characteristics,the 16S rRNA gene sequence analysis,isolated from the whole plant corn silage to extract 1 strain of Clostridium butyricum and the strain preparation for agents,added in the high moisture corn grain,by sampling and analysis,the strain can promote the butyric acid fermentation of high moisture corn silage.The primary results of this research were as follows:Test one:from the whole plant corn silage to extract 1000 strains of bacteria,through the physiological and biochemical identification,16Sr DNA sequence identification of non compliance with the suspected bacteria,the sequencing results submitted to the NCBI site for comparison.The results of the experiment showed that,by comparing the sequence results,3 strains were identified as Clostridium butyricum.Clostridium butyricum explore at different temperatures(0 ?,10 ?,20 ?,30 ?,40 ?,50 ?,60 ?,70 ?,80 ?),different pH values(pH of the liquid medium initial value is set as 3.5,4.0,4.5,5.0,5.5,6.0,6.5,7.0,7.5,8.0,10 gradient;the initial pH of the liquid medium was set to 7.0)under the conditions of the strain characteristics.When the culture temperature is 30 ? and 40 ?,the rapid growth of Clostridium butyricum;when the temperature is 0 ?,70 ?,80 ?,the Clostridium butyricum though slow growth,but can withstand low and high temperatures.At pH value of 6.5,7.0,7.5 Clostridium butyricum growth best pH value of slower growth,but the overall trend is when the pH value is low,rate of growth of Clostridium butyricum slower,reaches a certain concentration,the longer the time,at any time the pH value increased,the growth speed,reaches a certain concentration,the time shorter.It showed that this strain of Clostridium butyricum has a strong acid resistance,can be adapted to the acidic environment of the intestinal tract,and has good application value.The test results prove that strain characteristics through research,found that the species has a characteristic of high temperature and acid.Test two:the test points add amount of design and the high moisture corn grain material sterilization Group test for Clostridium butyricum;of high moisture corn grain to do different moisture Group;of high moisture corn grain do different crushing processing test.The feed additive was prepared from Clostridium butyricum,and added to the high moisture corn grain silage.The fermentation quality,the content of conventional nutrient and the stability of aerobic stability were analyzed.(1)On the high moisture corn grain for sterilization and non sterilization Group,adding different amounts of Clostridium butyricum,analysis of the various indicators of silage.The results showed that the fermentation quality and nutrient content of 1×106cfu/g of Clostridium butyricum were the best.(2)After drying the high moisture corn grain,the content of Clostridium butyricum with different adding amount was added,and the indexes of silage were analyzed.The experimental results showed that the initial water content of high moisture corn was 30%,the quality of silage fermentation and the nutrient content were the best.(3)High moisture corn grain in the pre silage to do a different degree of crushing Group,adding 1×106cfu/g of Clostridium butyricum,through the analysis of the nutrient content and other selection of the best degree of fragmentation of corn kernels.
