Clostridium chauvini infection,the pathogen of.which is Clostridium chauvini infection,is an acute,hot and septic infectious disease occurring in ruminants.Common typical symptoms are inflammation,gas swelling in the muscle plump parts of the affected animals,touch pressure twist pronunciation,the epidemic of the disease has a certain degree of land,seasonal,mostly in wet valleys,pastures,and in the hot rainy season.In recent years,there have been reports of bullishness and gangrene,which have brought great impact and economic losses to the development of animal husbandry and aquaculture,and also gradually threatened the safety and production of human beings.In order to meet the different clinical environment and detection needs,the suspected cases of emphysema were diagnosed quickly and effectively,and the epidemic and outbreak of the disease were controlled at the first time.In this study,20 positive samples were tested by four methods.The CctA gene with strong specificity was selected as the target gene for PCR detection,and the most suitable PCR detection method was established by screening the most suitable annealing temperature.An enzyme-linked immunosorbent assay(ELISA)for the detection of Clostridium perfringens was established by using Clostridium perfringens as antigen.the optimal working conditions of ELISA were determined,the optimal dilution was 1:200 and the optimal working concentration of ELISA was 1:2 000.Under the guidance of immunochromatographic assay and according to the characteristics of antigen-antibody,the developed colloidal gold immunochromatographic test strip for detecting goiter can be used for rapid and simple detection of goiter without adding any reagent.In the dot-ELISA method,the guinea pig was immunized with the most specific purified antigen by purifying the effective antigen extracted from the Clostridium perfringens standard strain,which was used as the primary antibody of the dot-ELISA method.The results showed that PCR,ELISA,colloidal gold test strip and dot-ELISA were successfully used to detect emphysema.the positive rate of PCR and colloidal gold detection method was 100%,and the positive rate of ELISA and dot-ELISA was 85%.Elisa and dot-ELISA have low specificity and high sensitivity,and are suitable for the detection of emphysema in epidemiological investigation.PCR method has high specificity,but has high requirements on experimental environment and conditions,and is suitable for laboratory detection.The colloidal gold test strip has good specificity,can be placed at normal temperature for a long time,is suitable for grass-roots units and clinical workers,and can quickly and efficiently detect a large number of samples.This study provides an important reference for the diagnosis of emphysema and provides a reliable basis for the prevention and control of the disease in the future.
