Taurine is the active substance that regulates the normal physiological activities of the body.It has the biological effects of maintaining the digestion and absorption of lipids,improving the immune ability of the body and enhancing the antioxidant capacity of the cell membrane.It also plays a certain role in piglet feeding and digestive tract function mediation.The aim of this study was to investigate the inhibitory effect of taurine intervention on LPS induced inflammatory activation in piglets.The intestinal injury model of piglets was established by intraperitoneal injection of lipopolysaccharide(LPS),and the inhibitory effect of taurine on inflammatory activation in piglets was studied.Through the level of serum immunoglobulin and inflammatory factors,the number of ileum SIgA plasma cell,the expression of ileum inflammatory factor mRNA,the expression of Toll like receptor TLR2,TLR4 mRNA and the expression of TLR4 signaling pathway in intestinal tissue,the effect of taurine intervention on intestinal immune barrier induced by LPS induced piglets was analyzed.24 weaned piglets were randomly divided into four groups for testing: control group,taurine group,LPS group,taurine + LPS group.The test period was 28 days.The taurine was fed evenly into the diet,the control group was fed with basal diet + injection of physiological saline,taurine group was fed with diet mixed with physiological saline and taurine + injection,and LPS group was fed with basal diet + LPS,taurine + LPS group was fed with taurine + LPS.The injection time was the twenty-eighth day.Specimens were slaughtered three hours after the injection,blood samples and ileum were tested,and the alleviation of taurine on acute stress was analyzed.Compared with the control group,the immunoglobulin(IgA,IgM,IgG)in the serum of LPS group was significantly lower than that of the control group,while the LPS+ taurine group was significantly higher than the LPS group(P < 0.05),and the serum inflammatory factor level(TNF-,INF-gamma,IL-6)in LPS group was significantly higher than that of the control group(P < 0.05),but the serum levels of the serum LPS+ taurine group were significantly lower than those in the LPS group(P < 0.05).The number of sIgA in ileum of group LPS was significantly lower than that of control group(P < 0.05),while LPS+ taurine group increased significantly compared with LPS group(P < 0.05).The expression of inflammatory factor mRNA in the ileum(TNF-,IL-6,IL-12)LPS group was significantly different from that of the other three groups(P < 0.05).The LPS group of Toll like receptor TLR2 and TLR4 was significantly higher than that of the other three groups(P < 0.05).The expression of p38 and phosphorylated LPS histone in the I kappa B alpha and MAPK signaling pathway in the NF-kappa B signal pathway of the TLR4 receptor in the ileum mucosa of piglets was significantly higher than that in the other three groups.ERK and its phosphorylated level of the LPS group was significantly higher than the control group and the taurine group(P < 0.05);but comparing the taurine + LPS group and the LPS group,although there is a downward trend,there is no significant difference(P > 0.05).To sum up,taurine intervention can inhibit the release of inflammatory factors induced by LPS induced immune stress piglets,control the inflammatory reaction and protect the intestinal immune barrier.
