In this experiment,the transformation and regeneration system of soybean cotyledon node and the genetic transformation system of soybean callus were established in 20 varieties of the main varieties of Northeast China.The regeneration rate of shoot,the rate of cluster bud resistance,the GUS dyeing rate of cotyledon node and the GUS dyeing rate of the callus were compared and studied.To select the most effective soybean varieties suitable for soybean genetic transformation test,to establish excellent soybean transformation system and soybean regeneration system for soybean laboratory in Northeast China,and provide the theoretical basis and data support for selection of varieties.The main results are as follows:1.In the selection results,Tiefeng 31,Shennong 20,Liaodou 13,Liaodou 32,Dongdou 17,Heinong 59,Heinong 60,Nannong 31,these two explants of Agrobacterium-mediated transformation test,all showed high conversion efficiency.2.Zhonghuang 35,Shennong 8,Liaodou 50,Dongdou 641,Heinong 54,Tiebeans 81,the above varieties in two explants of Agrobacterium-mediated transformation test,all showed low conversion efficiency.3.Compared with the transformation efficiency of different varieties,it was found that some varieties,different explants in the process of transformation,produced different results,Zhonghuang 13,Liaodou 15,Dongdou 9,Tiedou 37,Zhonghuang 35,Shennong 20.The conversion efficiency of these 6 cultivars is related to the type of explants selected in the experiment.4.Zhonghuang 35,Shennong 20,these two varieties showed false positives because of their chimera,which resulted in lower conversion rate in cotyledons,and high conversion efficiency in Agrobacterium tumefaciens-mediated test of callus.Zhonghuang 13,Liaodou 15,Dongdou 9,Tiedou 37,These 4 cultivars showed high conversion efficiency in the Agrobacterium tumefaciens mediated test,but the conversion efficiency was lower in the Agrobacterium tumefaciens-mediated test.The results can be used to provide advice and guidance for the selection of soybean varieties suitable for different explants regeneration tests.
