| Pseudorabies(PR)is one of the most important diseases causing severe damage to the pig industry.The disease caused a series of serious diseases such as obvious neurological symptoms,infertility,and mummified fetuses in weaned piglets,finishing pigs,and sows.Recent studies of herpesvirus-encoded miRNAs have shown that some herpesvirus-encoded miRNAs are involved in viral replication and even play a key role in the regulation of the virus.At present,there are very few relevant studies on pseudorabies virus-encoded miRNAs.The purpose of this study was to investigate the serum expression profile of miRNAs encoded by pseudorabies virus and its application in the differential diagnosis of vaccine immunization and infection.The sera of pigs in different stages of growth were extracted and the serum of vaccine immunized pigs at different stages of growth and the serum of infected pigs after immunization were mixed.Total RNA was extracted using a kit for serum RNA extraction.Quantitative comparisons of expression profiles of miRNAs encoded by different serum samples of pseudorabies virus using RT-PCR and real-time fluorescence were performed.Bioinformatics methods were used to analyze the regulation of pseudorabies virus-encoded miRNAs and their regulation.The effect of virus infection on the expression profile of the host miRNA.In this study,the serum expression profiles of pseudorabies virus miRNAs were determined based on the differences in the expression of miRNAs in serum and immune serum infected with pseudorabies virus.At the same time,the significance of the differences in miRNA expression profiles in different serum samples was compared and screened to identify miRNAs that could serve as molecular markers.In addition,the possibility of changes in miRNA expression profiles after vaccination and infection as a differential diagnosis was also analyzed in detail.Based on the miRNAs identified in the literature,we selected 37 pseudorabies virus-encoded miRNAs and synthesized their primers to study the potential role of pseudorabies virus-encoded miRNAs in the relative expression of miRNAs in infected pigs and vaccine-infected pigs.Screening miRNAs that can be used for the differential diagnosis of vaccine immunity and infection.This study selected the literature based on the JS-2012 strain of the latest study found that 37 miRNAs encoded by pseudorabies virus,and its real-time fluorescence quantitative PCR,according to the specific primers selected 10 miRNA expression profiles are as follows: 4 miRNAs Upregulated,six miRNAs were down-regulated.Differences in the expression of these miRNAs may play a particularly important rolein the replication of the virus.Our results suggest that miRNAs will have changes in expression levels after infection with PRV after vaccine immunization,which is of great significance for further study of miRNA function.By comparing the serum expression profiles of the vaccine immunization group and the infection group,the possibility of using miRNA-5 as a molecular marker for the differential diagnosis of vaccine immunity and infection was analyzed.Studies have shown that miRNA-5 can be used as a molecular marker for identification of vaccine immunity and infection.There will be more research on pseudorabies virus-encoded miRNAs.Studies in recent years have demonstrated that pseudorabies virus can encode miRNAs,and these miRNAs can also interact with the host.In the differential diagnosis of vaccine immunity and infection,based on the difference in miRNA expression profiles between two different types of serum,molecular identification is used as a molecular marker to identify vaccine-vaccinated pigs and infected pigs.The miRNAs screened in this experiment can be used as identification.The possibility of immunizing pigs with immune-infected pig markers was studied in depth.The results showed that miRNA-5 can be used to identify immunized pigs and infected pigs after immunization. |
