Soil salinization is a crucial factor that restricted the agricultural development.There are about 20%of land were affected by soil salinization.It is meaningful to explore the mechanisms of plant salt tolerance,improve plant salt resistance and then cultivate new salt-tolerance varieties.Salt-tolerant genes excavated and obtained is the key of molecular breeding for salt tolerance.ZTP29 gene?Zinc transporter?belongs to ZIP protein family,and it play an important role in balance the metallic ion of plant.In recent years,AtZTP29 was proved that has resistance to salt tolerance.Current research about plant ZTP9 are mainly concentrated on a few plants such as Arabidopsis thaliana?Arabidopsis Thaliana?,maize?Zea mays?,rice?Oryza sativa L.?.Cloning and function analysis of wheat?Triticum aestivum L?ZTP29 genes were reported rarely,research about wheat ZTP29 is still at the initial stage.In this study,the wheat TaZTP29 gene was cloned,expression pattern analysis was widely carried out under abiotic stress,subcellular localization of TaZTP29 were detected,and its function was analyzed through the transformation of model of plant Arabidopsis thaliana.The main results of this study are as follows1:The total length of TaZTP29 is 1032bp and the protein contained 277 amino acids with 8 protein transmembrane domain.The formula weight of TaZTP29 protein is 29.6 KD,and the isoelectric point is 6.30 belong to acidic protein.There is a conserved histidine residue in protein of fifth transmembrane domain.Evolutionary tree analysis indicated that TaZTP29 gene has homologous gene in monocotyledonous and dicotyledonous plants.2.Expression patterns under different stresses of TaZTP29 gene was investigated using real-time quantitative?qRT-PCR?assay.The results showed that:TaZTP29 gene was induced by salt stress and ZnCl2 solution of upregulated expression.Under the condition of high temperature and drought,the expression pattern of the TaZTP29 gene was down regulated.At grain filling stage,TaZTP29 gene expression was increased gradually,and reached maximum at 16 d after anthesis stage,after that time,the TaZTP29 gene expression reduced gradually.The results of tissue expression pattern analysis showed that the expression of:TaZTP29 was the highest in the root and the lowest in the grain.3.The subcellular localization results showed that the protein encoded by the 1:taztp29gene was on the plasma membrane of the cell.4.The results of salt tolerance identification showed that the salt tolerance of Arabidopsis thaliana transferred to wheat TaZTP29 gene was stronger than that of wild type Arabidopsis thaliana.The results showed that wheat TaZTP29 gene had salt tolerance function.
