Study On Artificial Insemination Using Sex Controlled Frozen Semen And Embryo Transfer In Dairy Goats

Abstract/Summary:
In recent years,the dairy goat industry has developed rapidly.Artificial insemination and synchronization of estrus technology have been used in intensive and large scale aquaculture to expand the genetic impact of excellent male and female goats.And the combination of superovulation and embryo transfer technology has made excellent species available.The breeding efficiency of female goats exponentially increases,which is the best technical measure to increase the efficiency of expansion of dairy goats.In order to increase the birth rate of female goats,the study used Xinong Saanen Dairy Goat as experimental object.Combined the use of sex-controlled frozen semen production,artificial insemination,superovulation and embryo transfer techniques.Preservation condition screening,X,Y sperm sorting,sex-controlled frozen semen production,embryo transfer,and other technical researches were conducted to investigate the practical effects of sex-controlled frozen semen artificial insemination and embryo transfer techniques to increase the reproductive efficiency of dairy goats,and to facilitate the promotion in production.Practical expansion technology.The main findings of this study are as follows:1.The study on the semen storage temperature in dairy goats found that the sperm motility in the 19°C and 4°C groups was significantly improved than that in the 36°C group within 12 h(P<0.05).When the preservation time exceeded 12 hours,the sperm viability of the 19°C group was significantly higher than the others groups.In addition,when semen is stored for 12h at different storage temperatures,it showed that sperm membrane integrity and acrosomal integrity in the 19°C group were significantly higher than those in the other two groups(P<0.05),which were 80.60±4.28%and 82.80±2.59%,respectively.2.In the XY semen sorting study,the results showed that the percentage of effective stained area and percentage of X spermatozoa was higher than that of the other two groups(32 uL and 36 uL when the amount of dyeing solution was 34 uL,which were 70.93±2.56%and 29.75±1.77%.However,there was no significant difference between the two groups(P>0.05).3.The effects of two different FSH on ovarian morphology on both sides of the goats were compared.In terms of ovarian length,width,height and volume respectively were2.42±0.28cm and 1.93±0.27cm,2.10±0.27cm and 1.83±0.29cm,1.98±0.26cm and1.63±0.34cm,9.88±3.73cm~3and 7.49±2.52cm~3,Shihuida FSH was superior and more stable to Ningbo Sansheng FSH(P<0.05).In the study of donor goats superovulation program,the two FSH on the ovulation point,the number of follicles and egg cells in the goats were found to be different.the group which made use of Shihuida FSH were better than the FSH produced in Ningbo Sansheng on the obtained ovulation spots,the number of follicles and egg cells(17.14±8.13,7.43±4.22,and 10.21±7.48),However,there was no significant difference between two groups(P>0.05).4.The result of estrus synchronization showed that the estrus efficiency of FSH treatment was significantly higher than that in the PMSG group(P<0.05),95%and 70%,respectively.There was no significant difference between the two treatment groups in the indicators of estrus time and estrus duration(P>0.05).Estrus time for donor and receptor goats found that the estrus time of donor goats was concentrated within 12~36h after plugs were withdrawn,and the estrus rate was highest at 24h after withdrawal suppository which accounting for 85.7%.The estrus time of the receptor goats was concentrated within 24 to 48hours after plugs were withdrawn,of which estrus had the most goats at 36 hours after plugs were withdrawn,accounting for 93.3%of all recipient goats.5.In the sex controlled frozen sperm insemination experiment,the average ovulation number of donor goats was 10.21±7.48.Pregnancy rate was 28.6%,number of available embryo was 5.Three recipient goats were transplanted and conception rates was 33.3%.The number of birth female goat was one,and the gender accuracy was 100%.In conclusion,the effective storage temperature of dairy goat semen is 19°C,and 34 uL of dyeing solution can improve the semen segregation effect of dairy goats.FSH can significantly increase the estrus rate of recipient ewes.Sexually controlled frozen sperm insemination combined with embryos receive sex after transplantation.Aligned with the correct offspring,successfully explored the possibility of high-efficiency propagation technology in raising the birth mother lamb rate,which laid the foundation for further production and application in the later period.
