In this study,we cloned one flavonol synthase gene(FLS)from grape hyacinth Muscari armeniacum,using PCR technique,named MaFLS2.Bioinformatics analysis showed that the length of MaFLS2 cDNA is 1152 bp,which contains one complete open reading frame with 1056 bp.It codes 353 amino acids,which belongs to hydrophilic proteins.NCBI analysis showed that MaFLS2 protein had typical DIOX_N and 2OG-FeII_Oxy structure domains,belonging to 2-oxoglutarate-dioxygenase protein families.Real time PCR showed that:(1)MaFLS2 gene is a tissue specific gene in grape hyacinths,and it had highest expression level in flowers buds starting to pigment in M.armeniacum while in Muscariaucheri 'White Beauty',its expression peak value appeared in roots.(2)At the different flowering developmental phases,MaFLS2 gene expressed highest in uncolored flower buds of M.armeniacum,whose flowers are blue,its expression pattern showed a overall tread of reducing first,increasing before decreasing again to its minimum level.But in M.'White Beauty',whose flowers are white,MaFLS2 gene presented a different expression pattern,which its expression peak value appeared in flower buds staying the middle period of its veraison,and it showed a general expression trend of increasing before declining.Besides,we constructed the interterence vector and overexpression vector of MaFLS2 gene to analysis the function of MaFLS2 gene preliminarily.This study provides certain basis to explore the function of grape hyacinths FLS gene in flower coloring.
