Currently,piglet white scour cause significant economic losses to the pig industry in our country.Conventional antibacterial drugs still face plentiful challenges,because of the following reasons:the numerous serotypes of pathogenic bacteria in piglet white scour,the effect of preventing theinfection use of vaccines is not ideal,the drug resistance of the bacterial strains is constantly increasing and the drug residues can severely threaten human health.Therefore,developing the veterinary drugs with high safety and difficult-to-resistance is an important effective approach to solve the current problems.Chihuang anti-dysenteric granules are natural veterinary-use compound drugs targeting at piglet white scour,developed by Inner Mongolia Huatian Pharmaceutical Co.,Ltd.independently.Objective:To evaluate the therapeutic effect of Chihuang anti-dysenteric granules for piglet white scour and its own toxicity through the research on the antibacterial effects of Chihuang anti-dysenteric granules in vitro and its acute toxicity test and sub-chronic toxicity test and experimental clinical trials.Further to determine the efficacy and safety of Chihuang anti-dysenteric granules and provide corresponding scientific references to the clinical application of Chihuang anti-dysenteric granules.Methods:Disk diffusion test and tube multi-proportion dilution method were adopted to determine the antibacterial activity in vitro,the minimum inhibitory concentration?MIC?and the minimum bactericidal concentration?MBC?of Chihuang anti-dysenteric granules.The toxicity of Chihuang anti-dysenteric granules was assessed through acute toxicity tests and maximum tolerance tests.The chronic toxicity of Chihuang anti-dysenteric granules was explored by sub-chronic toxicity tests.Artificial infection model of piglet white scour was established,and piglet white scour was treated with Chihuang anti-dysenteric granules to explore its experimental clinical treatment effect.Results:?1?The bacteriostatic zone diameter of Chihuang anti-dysenteric granules against enterotoxigenic E.coli was 15.22mm;the MIC of enterotoxigenic E.coli was125mg/ml,and MBC was 250mg/m L.?2?The results of LD50 determination test indicated that all the mice had no death during the test period,treated with once intra-gastric administration of 5.0g/kg BW Chihuang anti-dysenteric granules.What's more,the mice were energetic,acting normally,and no abnormal changes in food intake and water drinking.Thus,the LD50values were not determined yet.In the maximum tolerance tests,the mice were treated with5.0g/kg BW Chihuang anti-dysenteric granules for once intra-gastric administration and multiple intra-gastric administration within 12 hours.Within one week after given with the drugs,no abnormality was observed in the fur color and behavior of all the mice,and no death caused.After observation period,all the mice were put to death with cervical dislocation and anatomic observation showed no abnormal changes in internal organs.According to the maximum tolerance test of the mice,the maximum tolerance multiples to piglets was 130times.?3?During sub-chronic toxicity tests,all the male and female rats in the groups treated with high,medium and low dose or saline were orderly covered with fur,moved flexibly,defecated normally,and had no poisoning or death.Chihuang anti-dysenteric granules caused a significant difference in weight between the low-dose male rats groups and saline-treated male rats groups in the second week?P<0.05?.While the weight had no significant differences compared to other groups in other weeks.In terms of male rats,the weigh difference between high-dosage groups and moderate-dosage groups was significant in the first week?P<0.05?.From the second week to the fourth week where the experiments terminated,the weigh difference between moderate-dosage groups and saline groups,and low-dosage groups and saline groups were extremely significant?P<0.01?.The differences among high-dosage groups,moderate-dosage groups and low-dosage groups were also extremely significant?P<0.01?,while the difference between high-dosage groups and saline groups was not significant.This indicated that Chihuang anti-dysenteric granules had a certain elevation effect on the body weight of female rats,and the elevation effect of moderate dosage was ultra-significant?P<0.05?.The increase in male rats weight was not remarkable,and there were no obvious impact on the generation conditions,blood index,blood biochemical index,viscera index and the morphology of vital organs and tissues of rats.In this study,we successfully establish the disease model of piglet white scour.The treatment effect is best through administered with a moderate dose of Chihuang anti-dysenteric granules?20g/per pig?,and the effective rate and the cure rate were 100%and 80%,separately.The results above indicated that Chihuang anti-dysenteric granules has a higher safety in clinical medication.Conclusion:Chihuang anti-dysenteric granules possesses a good bacteriostatic effect on the enterotoxigenic E.coli,an major pathogenic bacteria which can lead to white diarrhea in piglets.This medicine itself has high safety and is a practically nontoxic drug.
