A survey was conducted on the Pice Mongolica in the Baiyinyubao Nature Reserve,1614 strains of endophytic fungi were isolated from different parts of healthy spruce by tissue fractionation.Different morphology was screened by observation of colony morphology and microstructure.The 195 strains of endophytic fungi were identified by morphologically binding molecular identification methods to determine the taxonomic status of the endophytic fungi population.Provide scientific basis for the species identification and resource utilization of spruce endophytic endophytic fungi,and for the next.One step further study of the endophytic fungi metabolites provides the theoretical basis for the following main results:1.The endophytic fungi in the Pice Mongolica are belongs to 27 taxa,22 species,14 genera,12 families,5 classes,namely Alternaria sp,Penicillium sp,Stachybotrysatras chartarum,Phoma sp,Aspergillus sp,Fusarium sp,Epicocum nigrum,Cladosporium herbarum,Curvularia aeria,Preussia sp,Chaetomium globosum,Cylindrocarpon sp,Epulorhiza sp Sydowia polyspora.It indicated that the endophytic fungi species of Pice Mongolica are rich in resources.2.Alternaria,Cladosporium,Fusarium,Phoma,Penicillium,Preussia organizations when the sand spruce parts all have distribution,mainly introduces for sand spruce,the endophytic fungi in groups from different organs of colonization rate,Alternaria and Penicillium for advantage,accounted for 49.7% of the total number of strains,36 ℅.3.After ITS-rDNA sequence analysis,endophytic fungi were mostly found in Ascomycetes and Deuteromycotina,corresponding to 4 classes and 10 families belonging to 12 genera and 22 species.Among them,Sydowia polyspora,a new record species,has not been reported yet.The Stachybotrysatras chartarum was first discovered in Pice Mongolica.4.From the perspective of the total separation rate of different tissue parts,the highest separation rate was shoots,followed by needles,again as cones,and roots and fruits were the lowest.The majority of endophytic fungi in spruce of Sandy Land show different degrees of specificity or preference of organs or tissues,while some fungal groups only occur in specific organs,such as Stachybotrysatras chartarum is an endemic species of fruit,and Epulorhiza sp only exists in the root tissue,and Sydowia polyspora is an endemic species of the skin.Overall,the population and quantity of endophytic fungi at all levels also showed a certain degree of regularity,ie lower> middle> upper Because the lower layer grows earlier than the upper layer,the growth time is longer than the upper layer,so the accumulation of endophytic fungi is greater in the lower layer,and the endophytic fungus also has richer biological diversity.This study on Pice Mongolica endophytic fungi separation identification and molecular phylogeny analysis,for the species identification of endophytic fungi of sand spruce and scientific basis was provided for the development and utilization of resources,and for the next step in-depth study provides the theoretical foundation endophytic fungi metabolites.
