| Gladiolus hybridus Hort is one of the four major cut flowers.The difference in dormancy of bulbs is the main reason that leads to uneven budding of bulbous bulbs,and there is a significant positive correlation between budding stage and flowering stage.The study found that low temperature treatment completely relieves dormancy.In order to reveal the changing trend and regularity of the key enzyme of organic acid,organic acid metabolism and reducing sugar in the dormancy release of bulbs,the bulbs of different gladiolus varieties at different times at 10°C were analyzed.The results were as follows:1.At room temperature(20-22 °C),Pink Pink Friendship and Red Supreme species did not break their dormancy after 60 days of putting on bulbs,but the dormancy began to break at 20 days at 10°C.Red Supreme completely broke the dormancy at 40 days,and Pink Friendship completely broke the dormancy at 60 days.2.The content of Ser in Pink Pink Friendship was significantly lower than that of Red Supreme except for 20 days in Pink Friendship.The contents of Ma and CA in the Pink Pink Friendship were significantly higher than those in Red Supreme,indicating that there were differences in Ser,Ma and CA contents between these two cultivar.The contents of Ile and Lys were significantly higher than the rest at 60 days.Except for 40 days,the content of Pro was significantly higher than that at 0 days.The content of GABA in Pink Pink Friendship increased significantly at 40 days and 60 days,and that in Red Supreme was significant at 60 days.The contents of Ma and CA were significantly higher than 0d in both varieties,as well as the content of Ma decreased after reaching the highest at 20 days,and the content of CA increased consistently.The results indicated that these organic acids are related to the breaking and breaking process of bulb dormancy.3.Except for 20 days,the activity of malate synthase in Pink Pink Friendship was significantly lower than that of Red Supreme.The activity of malate dehydrogenase was lower than that of Red Supreme,and there was a significant difference among 20,40 and 60 days,indicating that these two enzymes had differences between these two cultivar.During the whole treatment period,the activity of malate synthase showed a significant upward trend,and the activities of citrate synthase,γ-aminobutyrate transaminase,and phosphoenolpyruvate carboxylase increased significantly after 20 days and then decreased.The results indicated that these enzymes were related to the breaking of dormancy of bulbs.4.Xylose,galactose,ribose,and sucrose in Red Supreme and Pink Pink Friendship were significantly different throughout the time,indicating differences in the xylose,galactose,ribose,and sucrose varieties.During the whole treatment period,the content of fructose,xylose and ribose decreased significantly at 20 days and then increased significantly.Compared with 0 day,the contents of galactose,melibiose and sucrose showed a significant upward trend,indicating that fructose,xylose,galactose,Ribose,melibiose,and sucrose are related to dormancy breaking and breaking the process.5.Compared with 0 day,the ratio of sugar and acid significantly decreased and reached the lowest value at 20 days in Red Supreme,and then increased significantly.In Pink Pink Friendship,the ratio of sugar and acid significantly decreased and reached the lowest value at 40 days,and significantly increased at 60 days.It showed that the sugar-acid ratio was related to the process of dormancy breaking. |
