Soil salinization is one of the non-biological factors that restrict agricultural production and affect the ecological environment.The most efficient use of saline soil,increase of crop yield,increase of salt tolerance of crops,cultivation of salt-tolerant varieties,study of salt tolerance mechanism and identification of salt tolerance of crops have become current research hotspots.As an important grain crop and export product in China,red adzuki bean is rich in its resources and has great differences in salt tolerance,especially in the bud growth period,which is greatly affected by the soil environment.In recent years,research on red adzuki bean has mainly focused on drought resistance,functional characteristics and disease resistance mechanisms.Studies on the salt tolerance of red adzuki bean are rare,especially on the salt tolerance germplasm resources of adzuki beans.Screening and identification.The identification of the salt tolerance of other crops also has complicated processes and long measurement period.In order to explore the salt tolerance and optimum concentration and formula among different varieties of red adzuki bean,and to identify the easy method of salt tolerance of red adzuki bean,set different concentrations of 30 kinds of red adzuki bean to germination test.Statistical studies were conducted based on the measured indicators,and 30 red adzuki beans were sorted and classified.SSR molecular markers were used to verify the classification results.The experiment was conducted in the Institute of Life Sciences,Jiamusi University in 2017-2018.The water culture method used a distilled water matrix as a control group,and 32.5,65,97.5,130,162.5 mmol/L 5 concentration gradients were selected to screen and distinguish 30 species.The salt tolerance of red bean sprouts was determined by comparing the seed germination and agronomic traits of different treatments of red bean seed to the salt tolerance of the population,and the F value and the cluster analysis chart were used to analyze the salt tolerance of red adzuki bean seeds.Comprehensive sequencing and classification;using 60 pairs of SSR primers to identify the inheritance of 30 salt-tolerant germplasm of red soybean.The results of the study are as follows:1.By analyzing the relative indicators of the adzuki bean seed traits under the five salt concentrations,there are two results: one is that there is no significant difference in the indicators of adzuki bean seeds under low salt stress(?65mmol/L)salt stress.The high concentration(>65mmol/L)showed a downward trend under salt stress;the other was that with the increase of the salt concentration,all the indexes of the red bean seeds showed a downward trend.Through the correlation analysis of the measured indicators,it can be concluded that the root length and germination potential have the largest load in agronomic traits and germination characteristic factors respectively,so it can be used as the main identification index to screen the salt tolerance of red bean sprouts.2.The salt tolerance calculation formula was obtained by factor analysis,and the salt tolerance order was calculated by the formula using the germination index of 97.5 mmol/L each of the adzuki bean,and the cluster analysis of 30 red adzuki beans was divided into four categories according to the rank of salt tolerance binding.Group I included: Blush 9612;Group II included:Wild Adzuki bean,Blush 9218,Blush 12,Northeast Dahongpao,Longxiaodou 3,Pearl Red,Baoqinghong 7 varieties;Group III included:Jiahong1,Liaohong 8,Liaohong 4,Longkenhong,Nonganhong,Longxiaodou 2,Hongfeng8,Jihong 10,Japan red,Jihong 7,Jiahong 7,Pearl red,Baoan 947,Jilin red,Jihong 8,Magenta,Jihong 6,Xiaofeng 2,Liaohong1,Zhonghong 7,Jihong 9 twenty varieties;Group IV included: Tianjin red and Nongkenhong 2 varieties.Using Simple-sequence repeat(SSR),30 azuki bean germplasm resources were clustered and analyzed using cluster-averaging method and divided into four groups.The results of cluster analysis had a high correspondence with the classification based on salt tolerance.In summary,the following conclusions can be drawn from this test:1.The effect of salt solution treatment on the germination of 30 kinds of red adzuki seeds was divided into 2 groups.The low concentration of type I had no significant effect on the seed germination of 9 kinds of adzuki beans,and the high concentration inhibited the growth;the type II showed the inhibition of salt solution.21 kinds of red bean seed development such as pearl red.2.The 30 red adzuki bean varieties were divided into 4 categories under comparative analysis.Type I was salt-tolerant,Type II was moderately salt-tolerant,Type III was sensitive to salt,and Type IV was highly sensitive to salt.3.The germination index of 97.5 mmol/L red adzuki bean in 5 kinds of salt concentration can be used as the reference standard for evaluating the salt tolerance of adzuki bean.4.The salt tolerance of red adzuki bean can be preliminarily identified by the 97.5mmol/L salt tolerance formula.
