Arginine(L Arg)is required or mammals conditional necessary amino acid,plays a key role in embryonic development,and L-Arg young animals is the function of the essential amino acids,has important biological regulating function in body.It is well known that animals in the process of pregnancy,physical changes,such as increased blood viscosity,blood pressure,some blood clotting factors increase to show the change of blood rheology in high coagulation state and collective lowered immunity,etc.L-Arg can dilate blood vessels,and can significantly increase uterine placenta blood flow,improve the blood plasma viscosity and red blood cell quantity and deformation ability,so as to optimize the environment of embryonic growth and development,and Ca2+plays an important role in their metabolic regulation.Ca2+as the second messenger of the body,and is the indispensable material in various life activities,such as participate in nerve conduction,embryo implantation and development,energy transfer and gene expression regulation,plays an important role in the embryonic normal development.Verapamil as a regular Ca2+blockers can effectively inhibit extracellular Ca2+internal flow,thereby reducing intracellular Ca2+concentration.So determined on the basis of arginine can effectively relieve the IUGR explore the influence mechanism in the Ca2+during this process,the test by adding the optimal concentration of arginine in under the premise of fortified with verapamil as experimental contrast research embryonic development situation,and by measuring the weight different development of embryonic day age,corresponding to the placenta weight and female uterus weight to determine the L Arg and verapamil on embryonic IUGR,influence of using conventional paraffin section macrophages,and observation of mast cell count,using immunohistochemical method for microvascular endothelial cells and calmodulin counting positive cells were observed,enzyme-linked immunosorbent(ELISA)to explore the calcium dependent on the changes of nitric oxide synthase(nNOS)and eNOE measure in situ hybridization method to explore the mRNA expression of the experimental scheme,to illustrate the role of Ca2+in ease IUGR.The results of the study are as follows:1,the number of fetal rats arginine group,fetal rat survival rate,fetal rat body weight,and female uterus weight IUGR index is significantly higher than control group(P<0.05),verapamil group decreased significantly(P<0.05)or extremely significantly lower than that of arginine group(P<0.01).Uterine MC number 2,arginine group significantly(P<0.05)or very significant(P<0.01)lower than the control group and verapamil group;Number uterine macrophages arginine group than the control group significantly(P<0.05),verapamil group to the control group significantly(P<0.05),arginine and verapamil group difference was significant(P<0.01);Arginine group of uterus microvascular(MVD)count was significantly increased(P<0.05)in verapamil group significantly(P<0.05).3,L-Arg can make uterine placenta Ca2+dependent eNOS and nNOS increased the content of the overall trend(P<0.05),and after adding verapamil can make the content of eNOS and nNOS reduces total trend(P<0.05),but on a different day age will exist in small volatility,and the content of eNOS and nNOS in placenta was significantly lower than in the womb(P<0.05).4,the five kinds of optimization of Ca2+binding protein using immunohistochemical analysis,screening,positive cells expression was measured and the rate of change to select optimal binding protein,the sequence from high to low is:a calmodulin(CaM)and protein kinase C(PKC),calcium phospholipid binding protein AnnexinⅡ,calcium nerve phosphatase(CaN),Orail.And gene expression to determine the most optimal protein CaM,found mRNA expression of verapamil group was significantly lower than control group(P<0.05)or extremely significantly lower than that of arginine group(P<0.01).Above all:arginine can improve the female,for such and strengthen uterine weight,increase fetal rats weight,reduce fetal rats mortality aspects such as strengthen female reproductive ability,and can increase the macrophages and microvascular endothelial cells,strengthen uterine placenta and enhance content of eNOS and nNOS in Ca2+binding protein and gene expression,and verapamil can inhibit the promoting effect,show that Ca2+during pregnancy fetal play a key role in normal growth and development.
