The porcine epidemic diarrhea,caused by the porcine epidemic diarrhea virus is a serious viral infectious disease that is harmful to hog production.PED,All age groups of herds are susceptible to infection this disease,and the most severe to sucking piglets,performance characterized by severe diarrhea,vomit,dehydration and death highly.There are many prevent PEDV vaccines sold at the current markets,but PEDV still appear in swine flocks immunized using PEDV vaccine,Since 2010 to 2016 disease of PED cause a loss to farm every year,lead to most of pig farms lose faith in vaccine intervention effects of PED.And yet,oral inoculated with virulent virus and tissue inactivated vaccine obtain effect of controlling PED in clinic application,especially before a acknowledged and effective prevention PED vaccine appeared,pig farms takes America prevention measures of PED as lessons(oral inoculated with virulent virus)or the traditional prevention measures(inoculate tissue inactivated vaccine).And for this to research safety and protective for sow after oral inoculated with virulent virus and immunization tissue inactivated vaccine,it is of great significance for prevent and control PED to new-born piglet within seven days.This research established Fluorescence quantitative PCR method to detect the contents of PEDV.Can objective quantitative content of virus in the sample based on CT value of samples tested,the reliable theoretical data is offered for clinical use of oral inoculated with virulent virus and prepare tissue inactivated vaccine.By making gilt oral inoculated with conjugated of PEDV positive to explore sow postchallenge dynastic regularity of virus shedding and antibody rules.Use build fluorescence quantitative PCR to detection faeces of experimental pigs during diarrhea.The testing found that the faeces postchallenge days 2 and 3 virus copies amount pretty close challeng sample,the faeces clear decreased at postchallenge days 6 and become negative at postchallenge days 15.By Laboratory developed ELISA method for detection of PEDV antibody,to test six experimental pigs tracking the blood 13 times:Detection IgG antibody in serum clear increased at postchallenge days 11,attain maximum detection value of IgG and IgG antibody in serum at postchallenge days 20.Match up detection sows pathogen of feces and serum antibody after challenge,the resulting arrived at to make sow oral inoculated with virulent virus is better in during prenatal 15 d and 30 d,the sow postpartum IgA and IgG to reach a higher level and lower rates of virus shedding,to achieve the purpose of protection piglets,especially for some pig farm period for developing PED.In addition,tracking three pig farms immune PED tissue inactivated vaccine.The clinical effect observation and antibody detection result that the antibody level of serum samples antibody parameters of 228 sow and 68 newborn piglets,and 18 colostrum samples after immunity,serves as well protective effect and control the prevalence of disease at some level after immunity tissue inactivated vaccine.But growth trends of serum-antibody IgA and IgG inferior to oral inoculated with virulent virus.
