Bacterial Flora Analysis Of Canine Pyometra By DGGE

Objective: In order to reduce the cost of canine pyometra conservative treatment,improve the cure rate of canine pyometra conservative method and lay the foundation for clinical treatment of canine pyometra,the bacterial flora structure of bacteria in the pus of dog's uterus was analyzed with PCR-DGGE.Methods: First of all,the dog was generally examined through the clinical symptom.Then,the final 8 cases of dog uterus pus was diagnosed though laboratory testing and imaging testing.A surgery under aseptic conditions was carried out to remove the uterus and collect the intrauterine pus.And the bacterial total DNA was extracted from the canine pyometra.At last,with this template(bacterial total DNA),the 16 S r DNA V3 region were amplified with universal primers.And the sequence of the 16 S r DNA V3 region was analyzed with PCR-DGGE.Results: There were 9 dominant bacteria in the Canine pyometra,including Proteus mirabilis,Klebsiella oxytoca,Escherichia coli,Escherichia fergusonii,Klebsiella pneumoniae,Enterobacter cloacae,Enterococcus faecalis,Pantoea agglomerans,and Enterobacter hormaechei.Bacteria are different in each case in the canine pyometra,of which Klebsiella oxytoca,Escherichia coli and Enterococcus faecalis is the existence of each uterus in the pus bacteria.Conclusion: The bacterial flora structure in 8 dogs was different from one another,and the clinical symptoms and results of several tests were also different.Results showed that different degree of change in the canine pyometra flora structure would happen,because of the different clinical symptoms and the state of dog's body.This study laid the foundation for the analysis of canine pyometra flora in dogs.
