Quinolones?QNs?are a kind of synthetic antimicrobial compounds that are used to treat a variety of intestinal bacterial infections and respiratory infections in humans and edible produce animals such as cattle,pigs,turkeys and chickens.This study established a method for the determination of 27 Q uinolones,including,balofloxacine?BAL?,lomefloxacine?LOM?,enoxacine?ENO?,Tosufloxacin?TOS?,fleroxacin?FLE?,nalidixic acid?NAL?,ofloxacin?OFL?,ciprofloxacine?CIP?,marbofloxacine?MAR?,danofloxacine?DAN?,Gemifloxacin?GEM?,orbifloxacin?ORB?,norfloxacine?NOR?,sarafloxacin?SAR?,oxolinic acid?OXO?,flumequine?FLU?,sparfloxacin?SPA?,moxifloxacin?MO X?,enrfloxacin?ENR?,gatifloxacin?GAT?,pazufloxaxin?PAZ?,nadifloxacin?NAD?,difloxacin?DIF?,antofloxacin?ANT?and pipemidic acid?PIP?in edible tissues of Chicken?including muscal,egg,fat,liver and kidney?with LC-MS/MS for perfecting the multi-residue detection methods of QNs.Weigh samples for precision,the sample were extracted two times by 1%acetic acid that as the extraction solution,and then centrifugation after the merger of the supernatant,45?rotary evaporation to 1mL2 mL,the sample were evaporated to dryness and dissolved the residue with mobile phase.Analytical condition of LC-MS/MS:Using electrospray ionization multiple reaction monitoring mode monitoring,the quinolone were separated on Luna C 18 column,with mobile phase Acetonitrile-0.1%formic acid,the flow-rate of mobile phase was 0.25mL/min,and column temperature was maintained at 25?,the injection volume was 5?L,and stop time was 25 min.The results of the method for simultaneous determination of 27 QNs in dible tissues of study of C hicken are as follows:The total average recovery for sebum fortifled with 27 QNs at three levels were56.3%108.1%with accepetable intra-day 0.4%14.9%,in the sebum,the inter-day was21.3%for ENR,the others inter-day coefficient of variation of the remaining was between1.9%18.5%.The total average recovery for eggs fortifled with 27 QNs at three levels were58.4%109.3%with accepetable intra-day 0.8%15.2%,In addition to DAN's intra-day coefficient of variation was 15.2%,the remaining drugs were less than 15%,inter-day coefficient of variation between 3.8%and 19.1%.The total average recovery for muscle fortifled with 27 QNs at three levels were68.4%102.4%with accepetable intra-day 0.1%14.5%and inter-day 3.8%19.1%.The total average recovery for liver fortifled with 27 QNs at three levels were60.8%108.7%with accepetable intra-day 0.3%14.6%and inter-day 2.2%19.4%.The total average recovery for kidney fortifled with 27 QNs at three levels were60.9%113.0%with accepetable intra-day 0.5%14.9%and inter-day 3.4%18.9%.Limit of detection and Limit of quantification:0.31.3?g/kg and 0.62.5?g/kg in kidney,0.12.0?g/kg and 0.33.2?g/kg in sebum,0.12.0?g/kg and 0.34.0?g/kg in liver tissue,0.11.3?g/kg and 0.33.2?g/kg in muscle tissue,0.10.5?g/kg and 0.32.0?g/kg in egg.The results showed that the LC-MS/MS determination methods described in this invstigation was demonstrated good reproducibility,high sensitivity and specificity,while totally match the analysis on residus of QNs in edible animal tissues.
