Abstract:Eurycoma longifolia belongs to the family of Simaroubaceae,which is native of southeast Asia.It is the most precious officinal plants with great economic value and large demands in the markets.The quantity of wild resources are decreased sharply due to the excessive exploitation,which makes its introduction become difficult and burdensome.This study established an efficient and stable vitro regeneration technology of E.longifolia.Five phenotypic provenances were used to measure the phenotypic traits and the morphological characters of the seeds.Meanwhile,Seed propagation and cutting propagation were develop in this study,and the explants were taken form the seeds and the pathway of multiple bud shoots was carried out.The main results obtained from this study are as follows:1.There are same seed sizes and shapes(all oval)of the five geographical seeds from different climates,there are significant differences in the seed weight of the geographical provenances from different climatic and environmental conditions.The results show that the thousand seed weight of the five seeds is coastal provenance greater than island provenance,lowland provenance,mainland provenance and mountain provenance.The value of thousand seed weight is between 305.4 g and 316.2 g.2.The germination percentage have highly significant differences in geogenic provenances from different climatic and environmental conditions.Five germplasm resources begin to sprouting within 26d to 33d and germinate in 46 d.The germination rate of the seeds were 28%to 52%.The seeds of coastal provenance first to germinate and the germination rate was the highest,which reaching to 52%.3.The seed seeding of Eurycoma longifolia grows slowly.The survival rate of transplanting can reach to 70%.The buds shoots in 50 days after transplanting and the average height of seedlings can reach to 2025 cm in six mounths.4.The Effect of rooting powder for cuttage of Eurycoma longifolia Jack were not well,there are no significant difference between different rooting powder.GGR rooting powder h ave a relative high rooting rate,the rooting rate can reach to 40.23%.5.The technology of tissue culture and rapid propagation of Eurycoma longifolia were established.The optimal disposal of seeds was soaked in 5 hours under the room temperature.The germination rate can reach to 71.7%and had lower pollution rate of 25.0%under the condition of striped the inside and ouside episperms.Used 1/2 MS medium supplemented with 1.0 mg/L 6-BA,0.1mg/L IBA,0.5 mg/L GA3,50g/L source and 6.0g/L agar,PH 5.8 as shoot elongation medium,the induction rate was up to 100%;Used improved MS medium supplemented with 1.0 mg/L 6-BA,0.1mg/L IBA,0.7 mg/L GA3,50g/L source and 6.0g/L agar,PH 5.8 as proliferation medium,the growth coefficient was up to 4.60;Used improved MS medium supplemented with 1.5mg/L IBA,50g/L source and6.0g/L agar as root differentiation medium,the rooting percentage can reach to 93.0%.Well-rooted plants were transferred to non-woven bags containing peat soil,perlite,vermiculite(2:1:1,v/v)with above 90%.
