Melon,which has important commercial value,has been one of the most widely cultivated horticultural crops around the world.And it has been a kind of significant pattern plants which can be used to interpret the molecular mechanism of fruit ripening.It will provide a theoretical basis for the cultivation of new varieties by studying the regulation of fruit ripening.It will also provide new ideas for the regulation of fruit ripening network.In this study,two differential expressed genes,Cm Hsp83 and CmNAC34,which were chosen in the transcriptome analysis of 4 periods during fruit development and ripening,were cloned with full length c DNA by RT-PCR in Cucumis melon,respectively.And 5 CmHsp90 genes were identified in the wide-genome of melon using bioinformatics methods.Meanwhile,gene structure,phylogenetic analys is and distribution of conserved motifs of all thes e genes were researched.The result indicated that CmHsp90 gene family is relatively conserved both in structure and evolution.The expression pattern of Cm Hsp83 and CmNAC34 were detected by RT-qPCR.The results showed that the two genes were expressed in fruit,and the expression of CmHsp83 and Cm NAC34 peaked in 40 DAP and 30 DAP,respectively.Then overexpression and gene editing vectors were constructed.And their functions were analyzed during fruit ripening in melon by trans ient expression.The results suggested that Cm Hsp83 and CmNAC34 might promote fruit ripening.In the study of transformation of melon,overexpression vectors and gene editing vectors were transformed into melon by ovary injection method.8 and 6 seeds were positive in 72 CmNAC34 and Cm Hsp83 overexpressied seeds,respectively.The positive effic iency of CmNAC34 and Cm Hsp83 is 11% and 8.3%.And 10 and 12 seeds were positive in 72 gene edited seeds,respectively.The positive effic iency of Cm NAC34 and CmHsp83 is 13.8% and 16.7%.1 seed with Cm NAC34 gene editing vector was actually edited in 10 positive seeds and the efficiency of CmNAC34 gene editing mutation is 10%.
