Glycyrrhizin uralensis Frisch.,is a kind of perennial herb belonging to the Glycyrrhizae whose overground part is shrub-like and underground part is well-developed.It is widely distributed in hills,deserts and desert borders.Glycyrrhizin commonly used in the field of Chinese medicines and pharmaceuticals.Its dry stem and root are put into the medicine.And it is widely used in feed processing,tobacco processing,food and cosmetics processing and other industries.Due to the tolerance for drought,high salinity,and the preference for calcareous soils,Glycyrrhizin has an important contribution to environmental construction in ecology.In this study,Methylation sensitive amplified polymorphism(MSAP)technology was used to analyze the changes of DNA methylation levels in the stem organs and leaves of triennial Glycyrrhiza treated by high voltage electrostatic field during the seed period.For stem,indicating that the average levels of full-methylation,hemi-methylation,and total methylation of the triennial Glycyrrhiza in the experiment group after the electric field treatment are lower than those in the control group.And the total methylation levels decrease by 3.93%-15.87%,with an average decrease of 9.71%.The full-methylation ratio of the leaves of the three year Glycyrrhiza uralensis plant treated by electric field increased and the ratio of the hemi-methylation decreased.The total methylation level decreased by 9.53%-22.51% compared with the control group,with an average decrease of 14.97%.Which manifested that the treatment of high voltage electrostatic field weakens the degree of methylation of Glycyrrhizin.Meanwhile,It was found that the average content of glycyrrhizic acid in the HVEF treatment group is 10.72%,2.81% higher than that in the control group,indicating that the electric field treatment has a positive impact on the content of glycyrrhizic acid in the Glycyrrhizin.For the first time,a part of the DNA fragment of Glycyrrhiza Methyltransferase with 946 bp length was cloned in the experiment and its corresponding protein’s high-level structure was analyzed,providing a basis for physical technology in the cultivation of Glycyrrhizin.
