Physalis pubescens L.is an annual medicinal and edible plant belonging to Solanaceae.It was found in the introduction test that the seed had a dormant character and it was difficult to germinate directly,which has restricted its cultivation.This article mainly studies the dormancy type,dormancy physiology and methods of dormancy release of the Physalis pubescens seeds.The main conclusions are as follows:1.In order to determine the dormancy type of the Physalis pubescens seed,we conducted the stratification germination test,the seed coat permeability test,the alpha-amylase activity test and the biological identification of the germination inhibitor.The results showed that the water permeability of the Physalis pubescens seeds was good and the effect of the germination inhibitor was not obvious.The low temperature stratification at 4°C could significantly increase the seed germination and the activity of ?-amylase.2.Low temperature stratification can effectively promote the germination of the Physalis pubescens seed.In order to study the physiological and biochemical changes of seeds in the process of stratification,the seeds that were stratified and treated at different times were tested for their nutrient content,antioxidant enzyme activity,and hormone content.The results showed that the content of soluble sugar remained unchanged from 1.19% to 2.05% in the early stage,the starch content decreased from 4.21% to 1.96%,and the soluble protein content increased from 5.64 mg/g to 12.90 mg/g and then decreased to 6.24 mg/g,the change of nutrient content is closely related to the germination ability of seeds;the general trend of antioxidant enzyme activity is a gradual increase in the early stage of the stratification and the decline,which is consistent with the change of the germination ability of seeds,and the activity of antioxidant enzymes increased significantly during the stratification process,which can effectively remove the active oxygen produced by this process,and affect the germination of seeds.The content of auxin in seeds continuously increased from 3.62 ?g/g to 7.18 ?g/g,and the seed germination was promoted in the early low concentration;the content of endogenous gibberellin increased from 12.37 ?g/g to 19.39 ?g/g then showed a slight decrease,and it showed a promotion on the release of seed dormancy;the content of abscisic acid continued to decrease from 29.89 ?g/g to 12.69 ?g/g,showing the inhibition on the germination of the Physalis pubescens seed.3.The experiment was conducted to investigate the techniques of breaking the Physalis pubescens seeds dormancy by conducting seeds germination trials after soaking the seeds in solutions of potassium nitrate,hydrogen peroxide,gibberellic acid and polyethylene glycol.The results showed that,all different treatments significantly improved germination profiles of Physalis pubescens seeds.Seeds soaked in 1000mg/L gibberellic acid treatment had higher germination rate,germination potential and germination index than that treated with water(P <0.01),the germination rate increased from 14.67% to 41.33%.Effects of the treatment with 0.05% hydrogen peroxide soaking for 24 h was the second best,germination rate increased from 14.67% to 38%.Compared with single treatment,the interaction of hydrogen peroxide and gibberellic acid can only improve the germination potential of Physalis pubescens seeds,while other indices are not obvious.
