Dairy mastitis is a common disease of dairy cows.It has a great impact on the production of dairy cows,such as declining of milk yield and quality,affecting farm income,and harming the health of consumers.Therefore,it is necessary to find a solution through the study of disease-resistant genes.By studying the SNP of cattle,the experiment hopes to genetically improve the cattle,reduce the incidence of dairy mastitis,reduce the economic cost of raising cows,increase the production performance of dairy cows,increase the milk production,reducte the number of heads of dairy cows.And also hope to seek effective SNP at the genetic level to incidence of mastitis.In this study,research materials use DNA of 645 Holstein cows which are from Chuzhou first dairy farm in Anhui province.Using the method of direct sequencing,PCR-RFLP and SSCP were to test SNPs of IL-8、CXCR1、CXCR2、TLR2 and TLR4.Using the SPSS22.0 statistical software was to analyze correlation of the SNP and DHI.The main findings are as follows:In the study,seven SNPs were found in the IL-8,such as g.202 C>T、g.264 C>T、g 1892 A>C、g.1939 G>A、g.2103 C>T、g.2209 T>G、g.2222 G>A.g.202 C>T mutations were only analyzed because the seven SNPs were linked mutations.It was found that MY,PP and PY of the CC gene type were significantly higher than TC and TT gene type(P < 0.05).Eight SNPs were found in the CXCR1,such as g.295 C>T、g.337 T>C、g.341 G>A、g.369 T>C、g.646 G>A、g.652 G>T、g.739 C>G、g.823 G>A.After analysis of the sequencing results,g.646 G>A and g.823 G>A were selected to correlation analysis.It was found that FY,MY,PP and PY of AA gene type in g.646 G>A were extremely significantly higher than AG and GG gene type(P < 0.01).It was found that MY and PY of AG gene type in g.823 G>A were extremely significantly higher than GG gene type(P < 0.01).It was found that FY of AG gene type in g.823 G>A were significantly higher than GG gene type(P < 0.05).Three SNPs were found in the CXCR2,such as g.13783 G>A、g.13895.G>A 、g.14444 A>G 和 g.14476 T>C.It was found that FY,MY and PY of GG gene type in g.13783 G>A were extremely significantly higher than AG gene type(P < 0.01),and it was found that PP of AG gene type in g.13783 G>A were extremely significantly higher than GG gene type(P < 0.01)..It was found that MY and PY of AG gene type in g.14444 A>G were extremely significantly higher than GG gene type(P < 0.01).It was found that FY of AG gene type in g.14444 A>G were significantly higher than GG gene type(P < 0.05).It was found that FY,MY and PY of TT gene type in g.14476 T>C were extremely significantly higher than TC gene type(P < 0.01),and it was found that PP of TC gene type in g.14476 T>C were extremely significantly higher than TT gene type(P < 0.01)..One SNPs was found in the TLR2.It was g.10508 G>A.It was found that PP and FP of AA gene type in g.10508 G>A were significantly higher than AG and GG gene type(P < 0.05).It was found that PP and FP of AG gene type in g.10508 G>A were significantly higher than GG gene type(P < 0.05).Three SNPs were found in the TLR4,such as g.245 G>C、g.5987 A>G 和 g.9422 C>T.It was found that PP of CC gene type in g.245 G>C were extremely significantly higher than GC and GG gene type(P < 0.01).It was found that PP of GC gene type in g.245 G>C were extremely significantly higher than GG gene type(P < 0.01).It was found that SCS of AA and AG gene type in g.5987 A>G were extremely significantly higher than GG gene type(P < 0.01).It was found that MY of CC and CT gene type in g.9422 C>T were significantly higher than TT gene type(P < 0.05).It was found that PP of TT gene type in g.9422 C>T were extremely significantly higher than CC and CT gene type(P < 0.01).By detecting the polymorphisms of IL-8,CXCR1,CXCR2,TLR2,and TLR4 genes and their correlation with DHI production performance,10 mutation sites were detected: The g.202 C>T of IL-8;g.646 G>A and g.823 G>A of CXCR1;g.13783 G>A,g.14444 A>G and g.14476 T>C of CXCR2;g.10508 G>A of TLR2;g.245 G>C,g.5987 A>G and g.9422 C>T of TLR4.Among of them,three newly discovered sites that can be used as molecular markers for breeding cows are: g.202 C>T of IL-8;g.14476 T>C of CXCR2 and g.245 G>C of TLR4. |
