One disease that cows often get is mastitis,which can have a very big impact on dairy farming.Staphylococcus aureus is a common pathogen causing cow mastitis.Antibiotics are an effective method of treatment.However,due to long-term use,bacterial resistance to antibiotics.The main reason for drug resistance is the formation of biofilms by bacteria.Chinese medicine has a great effect on the treatment of mastitis in cows in recent years and does not produce drug resistance.This study explored the effects of ethanol extracts of perilla on the biofilm formation of Staphylococcus aureus and studied the regulation of its expression of the membrane gene icaA.Then,the ethanol extract of perilla is combined with doxycycline to reduce the formation of biofilm and whether it can achieve a reduction in synergy.To improve new methods for the treatment of cow mastitis1.This study examined the resistance of clinical isolates of Staphylococcus aureus from cow mastitis.Using the double dilution method to measure the MIC of multiple antibiotics,Staphylococcus aureus SA3 was found to be a multi-drug resistant strain.2.Extracted several alcohol extracts of traditional Chinese medicine,it then acted on the growth and biofilm formation of Staphylococcus aureus.As a result,it was found that the ethanol extract of perilla does not affect the growth of Staphylococcus aureus but can inhibit the formation of biofilm.3.Obtaining icaA gene related to membrane formation of Staphylococcus aureus by PCR.4.Staphylococcus aureus isolate SA3 electrotransferred into green fluorescent plasmid,observation of control group and non-adding SA3 biofilms by laser co-microscopy.As a result,it was found that the formation of biofilms in the ethanol extract of perilla,doxycycline,and combination group was significantly lower than that in the non-additive group.5.Ethanol extracts of perilla in combination with doxycycline to reduce biofilm formation and icaA gene expression.The results showed that the ethanol extract of perilla alone could inhibit the formation of biofilm and reduce the expression of icaA gene.Combined doxycycline can produce synergistic effect at a certain concentration to achieve the effect of reducing the synergy.In summary,this study use the ethanol extract of perilla on the Staphylococcus aureus biofilm,which reduced biofilm formation.The use of RT-qPCR confirmed that the ethanol extracts of perilla can reduce the expression of membrane gene icaA.In the case of a combination of doxycycline bioco-coat and icaA expression,synergistic effect can be achieved at a certain concentration to achieve the effect of reducing the synergy.Therefore,this study provides a new idea for the treatment of Staphylococcus aureus by Chinese herbal extracts.
