Aphis craccivora Koch and Galleria mellonella are important agricultural pests.Bean aphid can cause serious damage to leguminous plants and lead to a serious drop in the yield of legumes.Galleria mellonella main damage to the Apis cerana.The development of efficient and safe agricultural pest insecticides has great significance in practice.Tea and eucalyptus oil is mild in nature and has many activities such as deworming and insecticidal activity.However,there is not much research on the insecticide of tea and eucalyptus oil.At present,only the main component of tcrpinen-4-ol is found to have great killing Insect activity.However,the use of a single component tends to make pests resistant.In this paper,the insecticidal activity and insecticidal mechanism of five kinds of active ingredients of tea and eucalyptus essential oils mixed with specific emulsifiers on agricultural pests such as aphid and wax moth were studied.The effects of monomer insecticidal activity of terpinen-4-ol against aphid and wax moth and its Effect on Na+,K+-ATPase.The main findings are as follows:1 Terpinen-4-ol insecticidal activity of agricultural pestsTerpinen-4-ol had a strong contact activity against the wax moth,and had a strong insecticidal activity against the bean aphid,whether it was fumigation or contact treatment.The results showed that there was a significant difference in the virulence of wax moth and aphid with different dilution times of terpinen-4-ol.With the increase of the dosage of terpinen-4-ol,the mortality of the wax moth and the aphid also increased.The LD50 of terpinen-4-ol against the wax moth was 115.851 7μg/head.The LCso and LD50 values of terpinen-4-ol on aphid were 4.834 2 mg/L and 10.282 7 μg/head.2 Terpinen-4-ol insecticidal mechanism of agricultural pestsThe effects of terpinen-4-ol exposure on ATPase activity of Na+,K+-ATPase,Mg2+-ATPase,Cat+-ATPase,Ca2+,Mg2+-ATPase were measured.It was found that terpinen-4-ol had a strong inhibitory effect on Na+,K+-ATPase in wax moth.The contact toxicity of terpinen-4-ol with different dosage to the wax moth was significantly different,and the inhibitory effect was enhanced with the increase of terpinen-4-ol concentration.The effect of terpinen-4-ol on Mg2+-ATPase,Ca2+-ATPase,Ca2+,Mg2+-ATPase activity also has a significant inhibitory,but the inhibitory effect with terpinen-4-ol concentration does not have a significant positive correlation.Terpinen-4-ol has significantly reduced aphid Na+,K+-ATPase activity by treatments of fumigation and contact,when the dose of terpinen-4-ol increased,the inhibition enhanced.There was a significant correlation between dosage and inhibition.3 the composition of the essential oil and the best ratioIn this experiment,the main active ingredients of tea and eucalyptus oil were terpinen-4-ol,1,8-eucalyptol,a-terpinene,y-terpinene and terpinolene according to certain proportion and surfactant TritonTMX-100 mixed to obtain a compound essential oil.The preparation method is to mix these active ingredients according to a certain ratio into crude oil,and then mix with the surfactant TritonTMX-100 according to certain proportion.The volume ratio of the various active ingredient extracts is:Terpinen-4-ol:1,8-Eucalyptus:a-terpinene:γ-terpinene:terpinolene = 7:2.5:20:45.5:25;crude oil:TritonTMX-100= 3:1.4 compound pesticide insecticidal activity of agricultural pestsThe fumigant and contact activity of the compound essential oil with different concentrations on wax moth was studied.The results showed that the contact effect of the compound essential oil on the wax moth was significant.The contact toxicity of the compounding oil with different dosage to the wax moth was significantly different.With the increase of the treatment dose,the mortality of the wax moth also increased,The LDso value was 185.669 7 μg/head.The compound essential oil also had a strong fumigation effect on the wax moth,but the fumigation virulence of the compound oil on the wax moth was significantly weakened after the compounding oil was diluted.There was no significant correlation between the treatment dosage and the mortality of the wax moth.5 compound essential oil insecticidal mechanism of agricultural pestsThe effects of compound essential oil on the activities of ATPases such as Na+,K+-ATPase,Mg2+-ATPase,Ca2+-ATPase,Ca2+,Mg2+-ATPase were measured.It was found that after the compound oil was exposed to the wax moth,the activities of Na+,K+-ATPase and Mg2+-ATPase were significantly inhibited,and the inhibition of Na+,K+-ATPase activity by different doses of essential oil Significant difference.Withthe increase of compound essential oil concentration,the inhibitory effect was enhanced.The activity of Ca2+-ATPase was significantly inhibited.The difference of Ca2+-ATPase activity in different dosage of essential oil was not significant;Ca2+,Mg2+-ATPase activity was significantly inhibited,but there was no correlation between the intensity of the inhibition and the treatment dose.
