Selenium has a series of biological function which were very important,for example,the regulation of body's metabolism,to enhance immunity and disease resistance,to promote animal breeding,anti-tumor and anti-oxidation.Chitosan is an important biological macromolecular substance in organism,which could strengthen the function of immunity,anti-aging,anti-oxidation,anti-cancer and anti-rheumatism.We can obtain carboxymethyl chitosan through the method of modification of chitosan structure transformation,which could improve its antiviral activity and the immunity.Carboxymethyl chitosan selenium is the organic compounds of carboxymethyl chitosan and selenium,could play a dual function of selenium and chitosan.In order to research on the antioxidant function and molecular regulation mechanism of carboxymethyl chitosan selenium,We compound carboxymethyl chitosan and sodium selenite,which will provide a scientific basis for its wide application.also will provide a scientific basis for its application,The main results were as follows:1.In order to optimize the synthesis conditions of carboxymethyl chitosan selenium,the experiment of different doses of carboxymethyl chitosan,sodium selenite,acetic acid and reaction time were studied with single factor experiment,We use the influence factors to get the optimum reaction conditions by orthogonal test.The results of single factor experiment suggest that carboxymethyl chitosan,sodium selenite and glacial acetic acid these three factors in the effective conversion rate of selenium and selenium concentration are excellent.The results of orthogonal test indicated the optimum conditions are as follows: weigh accurately 1.25 g carboxymethyl chitosan was dissolved in 50 mL of deionized water,and then added the 0.1m L 1% glacial acetic acid in it,the sodium selenite 1.67 g dissolved in 10 mL of water added into carboxymethyl chitosan solution,after the 2h reaction,adding 180 m L ethanol,reaction at normal temperature,the edge suction side by ethanol washing 3 times on the next day,freeze drying to obtain the carboxymethyl chitosan selenium sample.Carboxymethyl chitosan selenium synthesis of amorphous white powder,odorless,selenium content was 109.3943mg/g,the yield was 39.49%.Through the ultraviolet spectrum and infrared spectrum analysis,confirmed the successful synthesis of carboxymethyl chitosan selenium.2.This experiment was conducted to research the effects of antioxidant function and immunity function of carboxymethyl chitosan selenium in mice.180 Kunming mice were randomly divided into three groups,60 rats in each group,half male and half female.including two treatments and one control.Mice in group III were fed with basal diet as control,while mice in group I and II and were fed with the basal diet supplemented with selenium as carboxymethyl chitosan selenium and sodium selenite(which calculated as the contains of selenium was 0.20mg/kg),respectively.On day 28 of the experiment,blood selenium concentration,glutathione peroxidase(GPx)activity,plasma superoxide dismutase(SOD)activity,malondialdehyde(MDA)content,and Con A-induced splenocyte proliferation were determined,and plasma interleukin-2(IL-2)and interferon-?(IFN-?)concentrations,splenic plaque-forming cell(PFC)responses,serum hemolysis level(HC50),and delayed-type hypersensitivity(DTH)responses were determined on day 15 of the experiment.The results showed that blood selenium concentration,GSH-Px activity,splenic PFC response,and plasma IL-2 and IFN-? concentrations in group I were higher than those in the group III and group II(P<0.01 or P<0.05),respectively.Plasma SOD activity,Serumhemolysis level,DTH responses,and Con A-induced splenocyte proliferation in group I were higher than those in control group(P<0.01 or P<0.05).Plasma SOD activity,serum hemolysis level,DTH responses,and Con A-induced splenocyte proliferation in group I were also higher than those in group II,while there was no significant difference between group I and group II(P>0.05).Plasma MDA content in group I was lower than those in the control group and group II(P<0.01 or P<0.05).Conclusion: carboxymethyl chitosan selenium in improving the blood selenium content,antioxidant capacity,cell immunity and humoral immunity,and the biological activity was better than sodium selenite.3.This experiment was conducted to study the Effect of Carboxymethyl Chitosan Selenium on Blood Selenium Content and Serum Antioxidant Indices Performance of Rabbit.Thirty six meat rabbits,average body weight was 2 kg,were randomly assigned into three groups,each group had two replications,every replication had 12 rabbits(male and female in half).Rabbits in group III(control group which calculated as the contains of selenium was 0.10mg/kg)were fed with basal diet,group I and II were fed with Carboxymethyl Chitosan Selenium and sodium selenite on basal diet(which calculated as the contains of selenium was 0.30mg/kg).The experiment lasted for 30 days.Took the blood of meat rabbit with empty stomach in the 10,20 and 30 day.The results showed that supplemental Carboxymethyl Chitosan Selenium increased selenium contents of blood and the activity of GSH-Px?CAT and T-AOC value in serum,decreased the activity of MDA in serum,compared with Sodium Selenite group.Serum activity of GSH-Px and T-AOC in group I had significantly different with group II and III(P?0.05)in the 20 day.Selenium contents of blood and serum activity of T-AOC in group I had significantly different with group III(P?0.05),serum activity of CAT in group I had significantly different with group II and III(P?0.05)in the 30 day.The results indicated that supplemental carboxymethyl chitosan selenium on basal diet of rabbits can increase blood selenium content and the serum antioxidant capacity of rabbit.4.This reseach was designed to explore the feed additive of carboxymethyl chitosan selenium on rabbit production performance,tissue antioxidant effects of selenium content and organization.36 rabbit were randomly divided into three groups,Group I and Group II were fed in Carboxymethyl Chitosan Selenium and sodium selenite on basal diet(which calculated as the contains of selenium was 0.30mg/kg),Group III was the control group fed the basal diet(which calculated as the contains of selenium was 0.1mg / kg),every replication had 12 rabbits(male and female in half),The experiment lasted for 30 days.Weighting the rabbit's weight at the beginning and end of the test.At the end of the test,take the rabbit liver,kidney,muscle,spleen and heart tissue,observed carboxymethyl chitosan selenium on rabbit production performance,tissue selenium concentration and antioxidant capacity of the affected tissue.The results showed that,compared with the control group,rabbit fed additive selenium source can significantly improve Rabbit average daily gain(P<0.05),Feed conversion ratio tended to decrease;which,carboxymethyl chitosan selenium Average daily gain was higher than selenite group,the former-feed ratio is lower than the latter;carboxymethyl chitosan selenium can increase the rabbit liver,kidney,muscle,spleen and heart tissue selenium content;add carboxymethyl chitosan selenium can increase rabbit liver,kidney and muscle GSH-Px,CAT,T-SOD activity and T-AOC,reduce MDA activity.The results described that in the rabbit feed additive carboxymethyl chitosan selenium can improve the performance of rabbit production,tissue selenium content and antioxidant capacity of the organization.
