Abstract/Summary:
Haemonchus contortus is a blood sucking gastrointestinal parasite of sheep and goats.Infection with this parasite can cause anaemia,weight loss and in some cases death.With increasing drug resistance problem in strongylid parasites,identification of potential new control targets is a priority.However,the main reason for the prevalence of haemonchosis is the rapid propagation and strong adaptability.So the exploration of the functions of genes controlling reproductive processes provides a profound foundation for the development of new vaccines and drugs.The eukaryotic 26 S proteasome is an enormous multisubunit complex that degrades the majority of proteins in the cell under normal conditions.It has been found to be of critical importance to cell cycle and cell survival.The 26 S proteasome can be divided into two subcomplexes: the 19 S regulatory particle and the 20 S core particle.Eukaryotic core particles contain six active sites,three on each of its two central β-rings,two termed chymotrypsin-like cut preferably after hydrophobic residues and have their catalytic residues located on the β5 subunits.In addition,substrate binding sites for each of these catalytic β subunits are formed as a result of specific interactions of the catalytic subunit with one of its adjacent β subunits.The integrity of proteasome is fatal to its functioning.In parasitic protozoa,the study of the function of proteasome has made some achievements,but the research in worms is still a blank.Many reports indicated β5subunit as the “rate-limiting” factor for the determination of total 20 S proteasome level.Therefore,I explored the function of β5 subunit of H.contortus by analyzing the localization of Hc-PBS-5 in H.contortus adult worm,inhibitor asssy by using borterzomib(BTZ)which was found to inhibit the chymotryptic activity of the proteasome by binding through its boronic acid residue to the beta 5 subunit of the core particle in S.cerevisiae,humans and E.multilocularis and by RNA interference in H.contortus.Results are as follows:(1)The characterization of Hc-PBS-5 sequenceMolecular and bioinformatic methods were employed to characterize proteasome subunit beta type 5 encoding gene.The alignment of amino acid sequence of Hc-PBS-5with homologues from different species showed conserved motifs and domains including Ntn-hydrolases signature motifs,metal ion binding sites,specific inhibitors binding sites and chymotryptic activity sites.The terminal of Hc-PBS-5 sequence contains hydrophilic amino acids making the protein molecular surface possessing the weak hydrophobicity and strong hydrophilicity.In addition,Hc-PBS-5 has the β-sandwich structure typical for Ntn-hydrolases by SWISS-MODEL prediction tool.The results of subcellular localizationindicted Hc-PBS-5 belongs to a kind of intracellular protease.No signal peptide and transmembrane regions were found in Hc-PBS-5 sequence,indicating that Hc-PBS-5protein is neither a membrane receptor nor a secretory protein.(2)The localization of Hc-pbs-5 in H.contortusRecombinant protein was expressed in Escherichia coli and polyclonal antibody against recombinant Hc-PBS-5 was produced by immunizing rabbits with recombinant Hc-PBS-5.Then immunofluorescence technique was used and revealed that Hc-PBS-5was expressed in the embryo cells of the egg and intestine,reproductive organs and tissues of the adult worms of H.contortus.(3)The efficacy of Boterzomib treatment in vitroIn proteasome chymotryptic activity assay,the extracts of x L3 Larvae demonstrated that BTZ display high anti-chymotryptic activity in H.contortus with an IC50 of 0.6 μM.The eggs were treated with BTZ in vitro,the hatching rate of the treated eggs slowed down in the low concentration inhibitor treated group,and the hatchability of the high concentration inhibitor treated group decreased.The survival rate of larvae in BTZ treated group was significantly lower than control group.In addition,there were many abnormal worms in treated group and less x L3 larvae developed to the L4 stage in BTZ treated group than in control group after culturing in vitro for seven days.(5)Silencing Hc-pbs-5 by RNAi soaking methodSoaking exsheathed-L3 in medium containing dsRNA of Hc-pbs-5 resulted in a significant decrease in the abundance of the Hc-pbs-5 gene transcript,as measured by quantitative PCR.During the initial 24 h exposure to the dsRNA,the gene suppression effect was quite specific to the targeted gene.Seven days after dsRNA treatment,the motility of treated x L3 was decreased(Not calculated)and the development of x L3 to L4 reduced 52.42% compared with the control group.The survival rate of treated x L3 also decreased significantly.
