Laparoscopic surgery has been preferred in many diagnostic and therapeutic processes in veterinary medicine,but laparoscopic pneumoperitoneum could result in liver IRI.The mechanisms of IRI included produce ROS,cell apoptosis and inflammatory cytokines release.Hydrogen could reduce their levels and protect organ injury induced by IRI.The aim of this study was to identify whether hydrogen could protect liver injury induced by laparoscopic pneumoperitoneum.Forty-five Wistar rats were divided into three groups?n=15?.The control group?C group?was subjected to anesthesia for 90 min.The experimental group?P15 group?was subjected to CO2pneumoperitoneum with 15 mmHg for 90 min.The intervention group?H2P15 group?was subjected to CO2 pneumoperitoneum with 15 mmHg for 90 min after injection with hydrogen gas?0.2 mL/kg?for 10 min.Rats were killed at 2 h,6 h and 24 h after surgery in each group?n=5?.Serums and liver tissue homogenate were detected liver function,oxidative stress markers and inflammatory cytokines.Some liver samples were stained with HE for histological studies.Some liver samples were detected the mRNA and protein expressions of Nrf2,HO-1,NQO1,Bcl-2,Bax,Caspase-3,TNF-?,ICAM-1 and IL-10 using real-time PCR and western blot.Compared with C group,the AST and ALT levels in serum of P15 and H2P15 group were increased at 2 h and 6 h.But compared with P15 group,their levels were significantly decreased in H2P15 group?P<0.01?.Their levels in liver tissue were similaras serum.Compared with C group,serum MDA was significantly increased in P15 group.But compared with P15 group,it was significantly decreased in H2P15 group?P<0.01?at 6 h.Compared with C group,serum SOD and GSH were significantly decreased in P15 group.But compared with P15 group,SOD was significantly increased in H2P15 group?P<0.05?at 2 h,GSH was significantly increased in H2P15group?P<0.05?at 6 h.Their concentrations in liver tissue were similar as serum.Compared with P15 group,Nrf2,HO-1,NQO1 and IL-10 mRNA expression were significant increased?P<0.01?in H2P15 group,Bax/Bcl-2,Caspase-3,TNF-?and ICAM-1 mRNA expression were decreased?P<0.01?in H2P15 group,the results were confirmed by western blot or ELISA.The experiment concluded that hydrogen could protect against liver injury during laparoscopic pneumoperitoneum through inhibiting oxidative stress,cell apoptosis and inflammatory cytokines release.
