| Southern rice black streaked dwarf virus(SRBSDV),belongs to Fijivirus Reoviridae,is persistent transmitted by white-backed planthopper.This virus was epidemic from 2009 to 2014,and resulted in severe yield loss in Southern China and South-east Asia.The rice yield loss has been lowered to 5% caused by this virus through national joint-vector prevention.However,the area and ratio infected by SRBSDV progressively raised and re-epidemically occurred in some local regions in Southern China.So supervising this virus is still pivotal for assessing its epidemic tendency.The pathogenic properties,transmission routes,symptoms,functions of a few encoded proteins,and viral genome structure of SRBSDV have been well-defined.However,the pathogenic mechanisms of SRBSDV and the molecular mechanisms of host resistance are still unclear.So,the research results of interaction mechanism between SRBSDV and the host at the molecular level would scientifically contribute to uncover the pathogenicity and assess the epidemic tendency of SRBSDV.The result of SRBSDV detection from rice samples in Hunan Province showed SRBSDV was still infected rice in Hunan Province from 2015 to 2017.Previously,SRBSDV Pns71 could interact with rice Cyns protein through yeast two-hybrid,colocalization and BiFC.In this study,the key domain(s)of Cyns interacting with Pns 71 was conducted to analyze interaction molecular mechanisms.Cyns gene contains two conserved domains,which high homology with RNA-binding superfamily and Cyanase-C superfamily,separatedly.The Cyns gene was divided to three domains and constructed into yeast expression vector.Y2 H result showed Pns 71 was interacted with the Cyanase domain of Cyns gene.For analyzing the biological functions of Cyns gene,the Cyns gene of rice and Nicotiana Benthamiana using VIGS(TRV system).The results showed silencing Cyns gene in rice resulted in death of rice.Nevertheless,Silencing Cyns gene in N benthamiana appeared mosaic white in leaves,similar symptoms of host infected by virus. |
