MSTN as a member of the transforming growth factor superfamily was discovered in 1997,which inhibit the expression of the gene that can cause the excessive proliferation of myoblast and muscle fiber hypertrophy,and cause skeletal muscle hyperplasia.Currently,studies have found that there kinds of natural mutant of myostatin in cattle,sheep,etc.,but pigs have not been found the natural mutation of myostatin.This experiment used TALEN gene editing technology and somatic cell nuclear transplantation(SCNT)technology to obtain the MSTN double mutation(MSTN-/-)and unilateral mutation(MSTN+/-)pigs.The main function of myostatin is to inhibit the growth and development of skeletal muscle,due to the biological function,makes its inhibitory factor or soluble receptor in promoting the growth of muscle.Inhibition of MSTN has been applied in the treatment of diseases such as Muscular dystrophy,muscle degenerative or metabolic diseases.Myostatin endocrine inhibitory factor including Follistatin,ActRIIB,Decorin and before the myostatin peptide,etc.A large number of research results show that these factors can promote the growth of muscle tissue and muscle cells.It is very effective to slow or inhibit type II diabetes and obesity by blocking the ability of myostatin.However,myostatin is not only expressed in muscle specific,but also in living cells of various animal tissues,and since its expression has affected the biological functions of these tissues or cells to some extent.Myostatin,for example,can control the structure and function of tendon tissue and adipose tissue metabolism myostatin has identified in mice with expression in breast tissue,besides expression in the muscles and breast tissue in the swine pituitary or rats is also expressed in the womb.Therefore,myostatin has a wide distribution in various tissues.In this experiment,we tested the MSTN gene Myostatin,Follistatin,ActRIIB,Decorin mRNA in two days of age MSTN-/-and MSTN+/-type and wild pig back longest muscle,heart,liver,spleen,lung,kidney,expression of the tongue.results showed that the expression of Myostatin gene in the heart and liver was MSTN-/-significantly higher than MSTN+/-and WT.In the spleen,lung,kidney and tongue,MSTN-/-,MSTN+/-pigs were all lower than wild-type pigs.The expression of Follistatin gene in the heart and liver is MSTN-/-significantly higher than MSTN+/-and WT,in the kidney and lung MSTN+/-significantly higher than MSTN-/-and WT,in the spleen and tongue MSTN-/-,MSTN+/-pigs are all lower than wild-type pigs.The ActRIIB gene is different in the heart and spleen,and there is no significant difference in liver,lung,kidney and tongue.The Decorin gene is different in the heart,liver,spleen,kidney and tongue,and there is no difference in the lung.Organ weight/weight MSTN-/-pig overall is smaller than MSTN+/-and wild-type pigs.In the liver and tongue,MSTN-/-pigs were significantly lower than MSTN+/-and wild-type pigs.In the lungs,MSTN-/-pigs were significantly lower than those in the wild-type pigs,and MSTN-/-pigs in the stomach were significantly lower than MSTN+/-pigs.Myostatin,Follistatin,ActRIIB and Decorin are different in MSTN-/-,MSTN+/-and wild-type pigs,and provide references for future studies on Myostatin,Follistatin ActRIIB,and Decorin genes in MSTN deletion.
