| Fragaria × ananassa is a perennial herb belonging to Fragaria genus in Rosaceae family.F.× ananassa is highly welcome for its rich in anthocyanins.Wild strawberry,as a kind of strawberry,is abundant in our country.In this study,we used Fragaria pentaphylla,Fragaria moupinens,and Fragaria nilgerrensisas research materials to reveal the metabolic differences and possible molecular mechanisms of anthocyanins in three wild strawberry varieties by using metabolomics,transcriptomics,quantitative PCR and HPLC technologies.In addition,F.× ananassa was used as the research material to study the expression mechanism of anthocyanin synthesis-related genes in different light conditions by using quantitative PCR and HPLC methods.The results provide the basis for further study of anthocyanin anabolic pathways and high-quality breeding of strawberry.1、Based on the metabolomics and transcriptomics data,1906 different genes were found significant difference expression between the red fruit of F.pentaphylla and F.nilgerrensis,of which 1363 metabolite up-regulated in red fruit of F.pentaphylla,the others were found down-regulated;1979 different genes were found significant difference expression between the F.moupinensis and F.nilgerrensis,of which 677 metabolite up-regulated in fruit of F.nilgerrensis,the others were found down-regulated;824 different genes were found significant difference expression between the F.moupinensis and F.pentaphylla,of which 399 different genes up-regulated in red fruit of F.pentaphylla.Most of those genes concentrated in plant secondary metabolism pathway.2、Quantitative real-time PCR analysis of the correlations between anthocyanin content and the relative expression of anthocyanin biosynthesis related genes in F.pentaphylla,F.moupinensis and F.nilgerrensis.The result showed that the expression of CHS,CHI,UFGT and MYB10 were significantly correlated with the anthocyanin content,MYB1 was significantly negative correlated.The correlation between flavonoid content and the expression of the F3’H gene showed a significant level.3、We determined the content of anthocyanin using HPLC and analyzed the relative expression of anthocyanin biosynthesis related genes using quantitative real-time PCR under the light transmittance of 100%,75% and 25%.The results showed that the content of strawberry under light transmittance 75% and 25% decreased 41.58% and 92.54%,respectively compared with the control.The relative expression of anthocyanin biosynthesis related genes and transcription factors,MYB10 and MYB1,also decreased.The expression of DFR,F3’H and UFGT were significantly correlated with the anthocyanin content.These results indicated that light was the key environmental factor affecting fruit coloring,and shading could inhibited the expression of anthocyanin biosynthesis related genes and transcription factors,blocked the anthocyanin synthesis in fruits,which finally contributed to the fruits coloring differences. |
