Fluorescent gold nanoclusters(gold clusters,Au NCs)have drawn great attention due to their good biocompatibility,unique optical properties.Au NCs have been previously reported in the field of biochemical detection,bioimaging,drug delivery and cancer treatment.However,the effect of Au NCs on virus is still an unpopular field.Therefore,the interaction of 2-mercaptoethane sulfonate(MES)and histidine stabilized Au NCs(His-Au NCs)were investigated,and the detection method of MES was established.Furthermore,the 2-mercaptoethane sulfonate and histidine stabilized Au NCs(MES-Au NCs)were synthesized through ligand exchange with His-Au NCs.Then,the influence of two types of Au NCs on the proliferation of PRV were researched.The specific research contents are as follows:1.This study have investigated the interaction of MES and His-Au NCs and discovered that the fluorescence of His-Au NCs will enhanced after reacted with the MES.Based on this,the detection method of MES was established.In addition,the results of UV-visible absorption(UV-vis),FTIR,fluorescence lifetime and X-ray photoelectron spectroscopy(XPS)analysis revealed that the formation of MES-Au NCs is through the ligand exchange between the MES and the histidine on the surface of His-Au NCs.This study not only established the analysis method of MES,but also provided new ideas for the detection of MES.2.The influence of His-Au NCs and MES-Au NCs on the proliferation of PRV were investigated through indirect immunofluorescence assay,plaque assay and western blot analysis.The results showed that the His-Au NCs can significantly inhibit PRV proliferation rather than MES-Au NCs.Further studies demonstrate that His-Au NCs inhibited the proliferation of PRV by inhibiting the process of replication,but did not affect the process of attachment,penetration and release of viruses.In addition,His-Au NCs did not directly inactivate PRV.Furthermore,cell imaging experiments revealed that His-Au NCs are distributed mainly in the nucleus,whereas MES-Au NCs are only distributed in the cytoplasm.The different localization of His-Au NCs and MES-Au NCs may be responsible for their various antiviral effects.Taken together,these results suggest that Au NCs have potential application value in the field of anti-virus and lay a foundation for the development of effective antiviral drugs.
