The hybrid breeding of rice takes a long time and is complicated to operate,and it is difficult to achieve the purpose of improving the traits in a specific gene.The use of CRISPR/Cas9 technology for site-specific editing of genes has been successfully used in rice.CRISPR/Cas9 technology was used to improve rice traits in this reserch.We selected wx,fgr,sd1,hd1,and ghd7.1 as candidate target genes,designed their CRISPR/Cas9 editing vectors,and constructed mutants of these vectors in different rice materials by Agrobacterium inoculation.By crossing or selfing,rice without T-DNA insert can be obtained.This method can quickly improve rice traits on the one hand,and on the other hand,it can also create mutants and provide new germplasm resources.The main results are as follows:1.According to the candidate gene design targets,the targets of the two candidate genes were simultaneously constructed into CRISPR/Cas9 editing vectors.Using Agrobacterium-mediated genetic transformation of rice seeds,transgenic plants of the T0 generation of H2613S-wx-fgr,Guizhou Xianghenuo-sd1-ghd7.12,Guizhou Xianghenuohd1-ghd7.11 and Xiangyubo-GS3 were obtained.2.Inbred and hybridized methods were used to obtain two gene homozygous functional mutant H2613S-wx1-fgr2,Guizhou Xianghenuo-sd1-ghd7.12,Guizhou Xianghenuo-hd1-ghd7.11 plants.3.To investigate the plant height,heading phenotype and other agronomic traits and quality traits of the T1 generation Guizhou Xianghenuo-hd1-ghd7.11,T1 and T2 generations of Guizhou Xianghenuo-sd1-ghd7.12,and to obtain reduced plant height.The Guizhou Xianghenuo improved materials with shortened growth period.4.The amylose content and aroma of T1 generation H2613S-wx-fgr seeds were detected,and the H2613 S improved material was obtained.In summary,this experiment succeeded in creating rice important mutants such as H2613S-wx-fgr,Guizhou Xianghenuo-sd1-ghd7.12,Guizhou Xianghenuo-hd1-ghd7.11,Xiangyubo-GS3,and improved target traits.With no change in production,no T-DNA insert materials are available,and new germplasm resources are provided for rice breeding.
