| Lapinized-attenuated Hog Cholera Vaccine(rabbit origin)is the main means to prevent swine fever because of its good immunogenicity and high security.But immunization failure of vaccine happen occasionally,the technological level and vaccine quality are also important reasons for immunization failure.The purification and filtration process can influence purity,efficacy and stability of Hog Cholera(HC)vaccine,impurities such as fat and protein can interfere with immune responses of vaccine,therefore influence immune effect.Objective:According to two main problems of many impurities and fat residual in the filter liquor existing in the production process of Swine Fever vaccine of a company,Hog Cholera live vaccine were purified by different purification techniques in this experiment.The aim is to standardize preparation procedure of Hog Cholera live vaccine(rabbit origin),and ensure vaccine quality.Methods:700 rex rabbits were inoculated with attenuated virus.Aftering determination of rabbit fever reaction,the qualified rabbits were killed,spleens and lymph nodes were asepsisly collected for the preparation of suspention.Experiment were divided into two groups:A group and B group.New patent filter(A group)and 80 mesh filter(B group)were used respectively to filter the same batch of suspention,then the filter liquors were detected as follows.1.The efficacy of semifinished product of lapinized attenuated Hog Cholera live vaccine(rabbit origin)were detected by efficiency test method for Hog Cholera live vaccine(rabbit origin)according to manufacturing instruction;2.Fat content in suspensions was detected by modified Babcock method;3.The particle size in tissue samples were measured used Mastersizer 3000E;4.The efficacy of product of lapinized-attenuated hog cholera live vaccine(rabbit origin)were detected by efficiency test method for Hog Cholera live vaccine(rabbit origin).Results:1.The efficacy of semi-finished product of lapinized-attenuated Hog Cholera live vaccine(rabbit origin)prepared by different purification techniques had no differences.Both can reach 1.8 X 104RID/g.2.Different purification techniques has no significantly effect on the uniformity of particle size(P>0.05).3.Different purification techniques has no effect on the fat content(P>0.05).4.The detection of efficacy indicated that the highest efficacy of vaccine can reach 1500RID per dose in both groups,Conclusion:There were no obvious influence on the efficacy of semi-finished product,uniformity of particle size and fat content of semi-finished product purified by different purification techniques,and the influence on efficacy of product was not significant too. |
