Microbial Strains Screening And Their Application In Rural Life Organic Waste Composting

At present,the rural environment problem has become the main obstacle of the new rural construction in China,the main characteristic of rural environmental pollution in our country at present stage is turning to the non-point source pollution from non-point pollution.One of the main factors of rural non-point source pollution is that the rural life organic wastes don't be disposed in a reasonable way.If the rural life organic wastes don't be disposed in a reasonable way,not only pollutes the soil and water resources but also wastes resources caused by the nutrients in the rural life organic wastes are not be used.Aerobic composting treatment has unique advantages as a rural life organic wastes treatment way compare with other ways such as incineration and sanitary landfill as well as anaerobic digestion.It not only can solve the staining problems cause by the rural life organic wastes but also can turn the rural life organic wastes into high-quality bio-organic fertilizer.And this paper points out the organic fertilizer broad market prospect in China by analyzing.The main research content of this paper is exogenous microbial additives using in aerobic composting.In other words the main contents of this study is screening of microorganism that can be used in aerobic composting through the separation and purification of strain and its research of enzyme production characteristics.And then the composite microbial agent by liquid fermentation and hybrid approach as equal ratio was obtained using these microorganisms.Then the effect of the microbial agent on fermentation process was tested by adding it to the composting system made up of fresh chicken manure and furfural residue.The results are as follows:(1)A amylase high-yield strain TY-40 was isolated.Its relative enzyme activity can reach 9.159U/mL,Three cellulase high-yield strains TY-3,TY-13,TY-40 were isolated.Their cellulase enzyme activity can reach 275.967U/mL,366.425U/mL,279.590U/mL,separately.One pectinase high-yield strain TY-27 was isolated.Their pectinase enzyme activity can reach 70.18U/mL.Among them,the stain numbered TY-1 has amylase activity and cellulase activity at the same time.(2)The antagonism test shows that the four strains isolated have no antagonism between each other.The 2L composite microbial agent by liquid fermentation and hybrid approach as equal ratio was obtained using these microorganisms.(3)The compost recipe was designed according to the fresh chicken manure and furfural residue indicator such as TOC,TN,moisture content before composting fermentation experiment.The best compost recipe was confirmed finally by calculated,the proportion of fresh chicken manure and furfural residue is 3:2,in this condition,the C/N in the composting material to 31,compost material moisture content was 55.6%.(4)To add homemade microbial agents into compost as treatment group,to add the same amount of sterile distilled water as blank control group,the two groups composting fermentation tests were carried out and the physical,chemical and biological indexes(such as pH,nitrogen content,dehydrogenase activity and GI)were determined in the process of composting fermentation.The experimental results showed that it can shorten the temperature rising time,improve the highest temperature,promote the organic matter degradation in material in compost material and speed up the compost maturity by adding manmade microbial agents into compost and furfural residue.It also showed a certain positive role in improving seed germination index,however,the two groups have no significant difference in terms of nitrogen fixation and deodorization.
