Hybridization technology is one of the commonly used techniques for genetic breeding of aquaculture animals.It also plays an important role in the breeding of abalone,and increasing the fertilization rate is the key technology.In this paper,the differences in fertilization rates under the combined effects of single factors and two factors in two important species of abalone cultured between Haliotis discus hannai?D?and Haliotis fulgens?F?were studied,and the appropriate conditions for hybridization fertilization were screened out,and also providing reference for the improvement of hybridization breeding technology between abalone species.The main findings are as follows:1.The effects of six single factors on the fertilization rates of DD,FF,DF and FD in four mating combinations of Haliotis discus hannai?D?and Haliotis fulgens?F?.?1?Water temperature:the suitable insemination water temperature for the self-reproduction combination?DD and FF?was between 20-22?,and the suitable insemination water temperature for the hybrid combination?DF and FD?was between 20-24 ?;?2?Salinity:the appropriate insemination salinity for the self-reproduction combination?DD and FF?was between 30-33,and the appropriate insemination salinity for the hybrid combination?DF and FD?was between 33-36;?3?pH:the suitable insemination pH of DD and DF was between 7.5-8.0,the suitable insemination pH of FF was between 7.5-8.5,and the suitable insemination pH of FD was between 8.0-8.5;?4?Sperm discharge time:the DD combination has a suitable insemination time within 2.5 h,FF combination within 1.5 h,DF combination within 1 h,and FD combination within 2 h;?5?Egg discharge time:the appropriate insemination time of DD combination is within 2 hours,the suitable insemination time of FF combination is within 3 hours,the suitable insemination time of DF combination is within 40 minutes,and the suitable insemination time of FD combination is within 50 minutes;?6?Sperm concentration:the optimal sperm concentration for the self-reproduction combination?DD and FF?is between 3.45×105 and 1.40×106ˇML-1,and the suitable sperm concentration for the DF combination is between 3.28×106 and 6.41×106ˇmL-1,and the suitable sperm concentration for the FD combination is between 1.02×106 and 6.41×106ˇmL-1.2.The effect of temperature and salinity interaction on the fertilization rate of DD,FF and DF in three mating combinations of Haliotis discus hannai?D?and Haliotis fulgens?F?.Two-factor experimental design was used to study the effects of interactions between temperature and salinity on the fertilization rate of the three abalone mating combinations,establish the fertilization rate model for each combination as follows?in the formula,each coefficient is the actual value?:DD Fertilization rate?%?=-1964.58 + 55.83*Temp + 90.39*Salt-0.06*Temp*Salt ?1.29*Temp2-1.36*Salt2FF Fertilization rate?%?=-1884.26 + 55.54*Temp + 84.13*Salt-0.03*Temp*Salt ?1.30*Temp2-1.26*Salt2DF Fertilization rate?%?=-1066.81 + 23.69*Temp + 52.68*Salt-0.09*Temp*Salt-0.48*Temp2-0.77*Salt2At the same time get the best insemination conditions for each combination:When the temperature was 20.76 ? and the salinity was 32.58,the DD fertilization rate reached a maximum of 87.34%and the satisfaction was 0.858.When the temperature was 20.94 ? and the salinity was 32.97,the FF fertilization rate reached a maximum of 84.32%and the satisfaction degree was 0.938.When the temperature was 21.35 ? and the salinity was 32.51,the DF fertilization rate reached a maximum of 42.53%and the satisfaction degree was 0.838.3.The effect of temperature and pH interaction on the fertilization rate of DD,FF and DF in three mating combinations of Haliotis discus hannai?D?and Haliotis fulgens?F?.Two-factor experimental design was used to study the effects of interactions between temperature and pH on the fertilization rate of the three abalone mating combinations,establish the fertilization rate model for each combination as follows?in the formula,each coefficient is the actual value?:DD Fertilization rate?%?=-2771.02 + 48.74*Temp + 589.01*pH + 0.16*Temp*pH-1.19*Temp2-37.14*pH2FF Fertilization rate?%?=-2772.63 +7579*Temp + 515.20*pH-0.04*Temp*pH-1.80*Temp2-32.04*pH2DF Fertilization rate?%?=-1641.21 + 32.70*Temp + 338.77*pH-0.51*Temp*pH-0.67*Temp2-20.75*pH2At the same time get the best insemination conditions for each combination:When the temperature was 20.92? and the pH was 7.98,the DD fertilization rate reached a maximum of 87.48%and the satisfaction degree was 0.831.When the temperature was 20.92? and the pH was 8.03,the FF fertilization rate reached a maximum of 87.80%and the satisfaction degree was 0.859.When the temperature was 21.25? and the pH was 7.90,the DF fertilization rate reached a maximum of 44.50%and the satisfaction degree was 0.868.
