Tea tree oil is an essential oil that has developed rapidly in recent years,it has broad-spectrum antibacterial activity.It has been developed into various types of products abroad,such as pre-servatives,antibacterial agents and fragrances.The antibacterial effect and mechanism of tea tree oil in China is still not fully understood.The research and development of its products lags behind.In order to promote the development and utilization of tea tree oil in China,the test study on the quality standard determination,antibacterial activity research,local irritation and studies of sensitization,acute oral toxicity,and subchronic toxicity,the result is as in the follows:?1?The tea tree oil quality standard was determined:the relative density of tea tree oil was 0.9011,the refractive index was 1.4815,the optical rotation was+8.83,and the pH was 5.46;the characteristic components of tea tree oil were qualitatively determined by TLC;GC-MS analysis was used to determine the content of the tea tree oil sign.The content of 1-8-cineole was 1.4%,and the content of terpinen-4-ol was 39.25%.?2?Tea tree oil antibacterial test:Tea tree oil is moderately sensitive to the antibacterial effects of the five test bacteria.The MICs of Escherichia coli and Staphylococcus aureus were 1.0%.The MICs of Staphylococcus aureus,Candida albicans,and Pseudomonas aeruginosa were 1.2%,1.4%,and 2.0%,respectively.Neutralizer identification test,neutralizer formulation:4.0%Tween-80+1.0%lecithin+2.0%glycine+PBS,can effectively neutralize tea tree oil residue on the test bacteria.In the suspension quantitative killing test,2.0%of tea tree oil killed Pseudomonas aeruginosa and E.coli for 20 minutes;4.0%of tea tree oil killed Staphylococcus aureus for 60 minutes;tea tree oil against white rosary.The killing concentration of the bacteria and Staphylococcus was 8.0%,and the action time was 60 min and40 min,respectively.?3?One complete skin irritation test:There was no edema and erythema in the four rabbits of the experimental group,and tea tree oil was not irritating to the white rabbit skin.A damaged skin irritation test:The low-dose group had no irritation to white rabbit skin;the high-dose group had mild irritation to white rabbit skin.Multiple complete skin irritation tests:In the experimental group,none of the four rabbits showed erythema,mild edema,no stimulation in the low-dose group,and mild irritation in the high-dose group.Acute eye irritation test:tea tree oil is not irritating to white rabbit cornea and iris,mild stimulation to the conjunctiva,can cause conjunctival hyperemia,but the symptoms are recovered after48 hours and are non-irritating.Local sensitization test:There was no skin erythema and edema in the test group.The skin reaction score was 0 and the sensitization rate was 0.?4?Acute oral toxicity test of tea tree oil:The LD50 of tea tree oil measured by BLiss method was2184.2 mg/kg,the 95.0%confidence limit was 1940 mg/kg to 2439.5 mg/kg,tea tree oil was a low toxic substance.Subchronic toxicity test of tea tree oil:ALT,AST and ALP content increased at the same time,TP content decreased,indicating that there is a certain liver and kidney damage in rats.The experimental group MCHC,PDW,P-LCR,MPV,TBIL significantly reduced?P?0.05?,indicating that the symptoms of anemia in rats;pathological slices showed liver and kidney edema in rats,the spleen has different degrees of congestion;The organ coefficient of the spleen of rats in the dose group was significantly increased.In summary,high concentrations of tea tree oil cause a certain degree of toxicity to the liver,kidneys,and spleen of rats,with no obvious toxic effects on the heart and lungs.This study confirms that tea tree oil has significant antibacterial activity,low toxicity,safety and non-irritating properties,it has a promising market research and development prospects.
