Upland Cotton(Gossypium hirsutum L.)and Island cotton(Gossypium barbadense L.)are allotetraploid cotton cultivars.G.hirsutum produce high yield variety,strong ability to adapt to the environment,the textile fiber quality,G.barbadense cotton production yield is low,a variety of environmental adaptability,textile fiber quality.Research on cultivation of upland cotton with the good quality of island cotton,breeding varieties with high yield,good quality,resistance to stress and suitable mechanization are the goal that breeders have been pursuing all the time.In this paper,the BIL(Backcross Inbred Lines)constructed with Zhong 36 and Hai 7124 as parents laid the foundation for the transfer of island cotton traits to upland cotton.The agronomic characters of BIL population were studied by SLAF-seq(Specific-Locus Amplified Fragment Sequencing),they are qualitative traits,petal color(Y1),pollen color(P1),petal sunred(PS);quantitative traits,chlorophyll content,leaf color.Metabolic and developmental mechanisms provide a useful tool.Quantitative traits such as yield and fiber quality are the primary objectives of breeders.In this paper,the primary mapping of qualitative and quantitative characters was carried out by using land and sea backcross inbred line population.The main results were as follows:1.According to the SLAF-seq(Specific-Locus Amplified Fragment Sequencing)results available in the laboratory,the BSA(separated egregant analysis)was carried out on BILs,according to the standard of Q value screening error rate less than 0.001(90%).A total of 12,388,465 reads were obtained from male parent,13,880,260 reads from female parent and 4,439,674 reads from offsprings.The petal color(Y1)was located in the three physical intervals of 22.58-29.09 Mb,41.24-48.85 Mb and 53.61-59.13 Mb on the chromosome A13;The anther color(P1)was located in the physical intervals of 39.80-40.64 Mb and 79.97-91.09 Mb on the chromosome A05,and the petals sunred(PS)was located in the physical intervals of 5.04-23.69 Mb on the chromosome D07 and the43.32-54.54 Mb on the chromosome D12.2.The QTL(Quantitative Character Loci)of chlorophyll content and leaf color were analyzed by laboratory map and ICIM(Inclusive Composite Interval Mapping)method.Nine QTLs of chlorophyll content were detected on 6 chromosomes,in which q-SPAD-A11-1 was detected at both stages.The phenotypic genetic contribution rates were 5.08%and 5.75%,and five QTLs were detected on 4chromosomes.The q-SPAD-10-1 mapped by chlorophyll content was located in the same position as the q-LCG-10-1 located on chromosome A10.Phenotypic genetic contribution rates were 5.28%and 4.71%respectively.Pearson correlation analysis between chlorophyll content and leaf color was carried out.The results showed that there was a significant positive correlation between leaf color depth and chlorophyll content.The QTL mapping of leaf color was reported for the first time.This study will be helpful to the fine location of chlorophyll content and leaf color and the molecular marker-assisted selection of cotton breeding with high light efficiency.
