This experiment was conducted to study the effect of different pH,AGD and VE additons on RNA/DNA and the expression of GPx and PPARa genes in marine medaka(Oryzias melastigma)larva.Select the 30d old,healthy marine medaka larva with the same size as research object.Using imported German AB salt mixture(davanced sea crystal)simulate artificial sea water.During the rearing period,water pH 8.1,temperature 28?,salinity 30,and dissolved oxygen was more than 6.0 mg/L.The experiments content and results are as follows:1.This experiment was conducted to study the effect of different pH on RNA/DNA and the expression of GPx and PPARa genes in marine medaka larva.The fish were randomly assigned into 5 groups with triplicate and fed basal diet for 6 weeks.The results showed that the ratio of RNA/DNA of marine medaka larva were significantly decreased by different pH,but the relative expression levels of GPx?PPARa genes were significantly increased.For instance,the RNA/DNA ratio of marine medaka larva(pH8.1)was 1.86±0.01,the 2-??Ct value of GPx?PPARa genes were 1.18±0.02,1.03±0.03,respectively.However,the RNA/DNA ratio of marine medaka larva(pH7.3)was 1.36±0.01 and the 2-??Ct value of GPx?PPARagenes were 1.71±0.05,1.52±0.04,respectively.2.This experiment was conducted to study the effect of different AGD and VE additons on RNA/DNA and the expression of GPx and PPARa genes in marine medaka larva at pH 7.7.The fish were randomly assigned into 6 groups with triplicate and fed for 6 weeks.Marine medaka were feed using one Control diet(GO)with no AGD and VE,and 5 treatment diets added different doses of AGD and VE.The results showed that the ratio of RNA/DNA and the relative expression of GPx and PPARa genes were significantly increased by adding different AGD and VE.For example,the RNA/DNA ratio of GO(adding AGD 0 g and VE 0 IU in Per kilogram of dry diet)was 1.65±0.01,the 2-??Ct value of GPx?PPARa genes were 1.60±0.01,1.43±0.04,respectively.However,the RNA/DNA ratio of G3(adding AGD 5 g and VE 50 IU in Per kilogram of dry diet)was 1.84±0.01,the 2-??Ct value of GPx?PPARa genes were 2.66±0.03,1.91±0.01.In conclusion:Ocean acidification can significantly reduce the ratio of RNA/DNA,induce the expression of GPx?PPAR ? genes.Adding different doses of AGD and VE can significantly improve the RNA/DNA ratio,increase relative expression of GPx?PPAR genes.In this experimental conditions,the kg of dry diet added AGD 5 g and VE 50 IU is the best optimal dose.
