Sugarcane pokkah beong was caused by Fusarium species complex,the mainly symptom of pokkah beong disease is leaf wilting,cane stems bent and deformed,internodes shortened,the worst is depraved in the headpiece of sugarcane,even death,However,bakanae disease of rice present Internode length because of gibberellins produced by Fusarium fujikuroi,in this paper,we used UHPLC-MS/MS,genomics,transcriptome to investigate the regulation of GAs metabolism of two species(Fverticillioides CNO-1,Fproliferatum YN41),Give the foundation for further research of the relation between pokkah beong disease and Fusarium's gibberellin metabolite.(1)According to UHPLC-MS/MS results,it turns out that CNO-1 has the ability of produce three GAs(GA3,GA4,GA7),the concentration of three gibberellins are 0.15.5.11.0.78ug/ml,respectively.However,YN41 cannot produce any GAs.(2)Alignment of gibberellin biosynthesis gene cluster in CNO-1 and YN41 reveals that CNO-1 contain the entire gene cluster,the seven genes are closely linked,which located in chromosome V,like CNO-1,YN41 also has the gene cluster in chromosome V,but missing an P450-4 gene,PCR test and sequence,alignment results are consistent with previous conclusion.(3)GAs detect in different medium shows that the concentration of three GAs(GA3,GA4,GA7)in fermentation medium are 1.97?5.12?1.34ug/ml,far higher than GAs in czapek's medium(0.03?0.31?0.29ug/ml).In this two medium,the concentration of GA4 all higher than GA3,GA7,especially in fermentation medium.However YN41 also can not produce any GAs in this two mediums.(4)Analysis of RNA-Seq for CNO-1 in different medium shows that seven key genes all significant down-regulated,accord with the GAs detect,qRT-PCR results also verified the RNA-Seq' correctness.However,in YN41,six key genes does not expressed or slightly expressed.
