| The sisimal cattle are bred in all countries of the world,and their varieties are divided into milk,meat and milk.Chinese sisimal bull is a kind of fine breed of cow,which has been introduced from Switzerland and has been improved by several generations.Chinese holstein cows(1997 years ago China piebold cows)is the end of the 19 th century,by a Chinese cattle with the introduction of Chinese holstein cows,after 1970 to 1980 years of breeding,and gradually formed.In this study,pcr-rflp was used to detect the genetic polymorphism of ABCA1 gene and further analyze the correlation between the SNP site of ABCA1 gene and the milk yield of Chinese holstein cows.Quinethiol is an important enzyme,which is an important enzyme that forms disulfide bonds,which is essential for protein folding and stability.Recently,QSOX1 was found to be involved in a variety of biological processes,including viral replication,embryonic development,inflammation and cancer.The structural integrity of the soluble protein dimerization has proved to have significant correlation with QSOX1,can be in bacteria through the human joint QSOX1 gene expression,a proper level of soluble protein dimerization in bacteria protein.In order to further verify the ATP combination box and A member of the family 1(ABCA1)gene and bovine fetal fibroblasts and the relationship between the Chinese holstein cows milk fat metabolism,improve the quality of milk in order to meet different people,this study examined the role of ABCA1 by RNAi technology.As far as we know,the role of ABCA1 has not been confirmed by RNA interference(RNAi).Therefore,we use FuGENE HD mediated transfection reagent RNA interference technology to reduce milk Chinese holstein cow mammary gland epithelial cells(BMECs)and fibroblasts(BFF)endogenous gene expression level,specific silence two genes,analyzing the change of the function of cell biology.In terms of mRNA and protein levels,fluorescence quantitative PCR technology,protein imprinting and tg-kit were used to analyze the content of triglyceride(TG)in cells before and after RNAi treatment respectively.The results showed that the mRNA expression levels of ABCA1 gene and QSOX1 gene were significantly decreased after RNA interference by fluorescence quantitative PCR technology,and protein levels were significantly reduced by western technology.Through the tg-kit kit,the expression of TG content in the cells was significantly reduced after the detection results showed that RNA interference.The results show that the ABCA1 gene and QSOX gene are important genes in the synthesis of triglycerides in the breast tissue and fibroblasts of Chinese holstein cows,and may be related to the synthesis of dairy cow’s milk fat rate.This study provides theoretical basis for building low-fat dairy cows. |
